Cefaclor-Induced Depression Model in Mice

To prepare the antibiotics-induced depression model, mice were allocated randomly in each group (n = 6/group). Mice were administrated orally with Cefaclor (200 mg/kg/day, dissolved in 0.2 mL saline) once a day for 5 days. Control groups were treated with vehicle (0.2 mL saline) instead of Cefaclor. The doses of Cefaclor used in the study were established according to their clinical doses65 and equivalent dose calculation based on body surface area66 (link). Cefaclor (PHR1283) was purchased from Sigma-Aldrich (St. Louis, MO).
To examine the antidepressant effects of fluoxetine on Cefaclor-induced anxiety- and depression-like behaviors in mice, mice were allocated randomly to each group (n = 8/group). fluoxetine (20 mg/kg/day, dissolved in 0.2 mL saline) was orally administrated once a day for 5 days67 (link). Control groups was treated with vehicle (0.2 mL saline) instead of fluoxetine. fluoxetine (PHR1394) was purchased from Sigma-Aldrich.

Quantification of Cefprozil Diastereomers

Chemical standards of cis- and trans-cefprozil (purity ≥ 98%; CAS No. 92665-29-7) were obtained from Hanmi Pharma, Co. (Seoul, Korea). Cefaclor (purity ≥ 99%; CAS No. 53994-73-3) was supplied by Sigma-Aldrich, Inc. (St. Louis, MO, USA). Acetonitrile (ACN), dichloromethane (DCM), methanol, and water (18.2 mΩ) of LC-MS/MS grade were also purchased from Sigma-Aldrich, Inc. (St. Louis, MO, USA). Ethyl acetate of HPLC grade was obtained from Fisher Scientific, Inc. (Hampton, NH, USA). In addition, LC-MS grade formic acid, acetic acid, and trichloroacetic acid (TCA) were purchased from Thermo Fisher Scientific, Inc. (Waltham, MA, USA). All chemicals used in this study had the highest HPLC grade or better quality available. Cefaclor was used as an internal standard (IS) in both methods. Fig. 1 shows structures of cis-cefprozil, trans-cefprozil, and Cefaclor.

Thiol-Modulating Reagents and Enzymes

Microcon cells and Immobilon-P Transfer Membrane were obtained from Amicon and Millipore, respectively. The bicinchoninic acid (BCA) kit for protein determination was from Pierce. Amoxicillin (Clamoxil) was from Beecham. Ampicillin (AMP), benzylpenicillin (BP), and cefuroxime were from Laboratorios Normon (Madrid, Spain). Deuterium oxide (D2O) for proton-nuclear magnetic resonance (1H-NMR) studies was from VWR. ECL was purchased from GE Healthcare.

HPLC Analysis of Cephalosporin Antibiotics

All reagents and solvents used were of analytical grade. Potassium dihydrogen phosphate (KH2PO4), di‐potassium hydrogen phosphate (K2HPO4), phosphoric acid (H3PO4), formic acid (HCO2H), HPLC‐grade acetonitrile (ACN) and methanol (MeOH) were purchased from Merck (Darmstadt, Germany). Water was purified by a Purelab UHQ system (ELGA, Marlow, UK). The reference standards of cefadroxil (CFD), cephalexin (CPL), cefaclor (CFO), cefixime (CFI), amoxicillin (AMO) and cefuroxime (CFU) were purchased from Sigma‐Aldrich (Singapore). The blank urine samples were collected from healthy children.

Antimicrobial Agents Characterization

Fifteen antimicrobial agents including penicillin, cephem, carbapenem, aminoglycoside, macrolide, tetracycline, quinolone, glycopeptide, and sulfonamide were used for this study (see Table 3). The following agents were purchased from Wako Pure Chemical Industries, Ltd. (Osaka, Japan): benzylpenicillin, cefazolin, piperacillin, ciprofloxacin, minocycline, and trimethoprim-sulfamethoxazole complex. Ampicillin, ceftazidime, ceftriaxone, cefaclor, gentamicin, erythromycin, and vancomycin were obtained from Sigma Chemical Co. (St. Louis, MO, USA). Amoxicillin was purchased from Fluka Biochemika (Bucks, Switzerland). Imipenem was kindly supplied by the Banyu Pharmaceutical Co., Ltd. (Tokyo, Japan).

Preparation and Characterization of N. sativa Powder

Seeds of N. sativa were purchased from the superstore, washed
to remove the impurities, air-dried, and crushed, and then the powder
was prepared by using an electric blender. To acquire uniform-sized
particles, the seed powder was sieved through a mesh. The obtained
powder was stored in an airtight jar for further analysis. The plant
material collected and used was according to the institutional guidelines.
The purchase of silver nitrate (AgNO3) and four antibiotics
(streptomycin, cefaclor, ciprofloxacin, and trimethoprim) was made
from Sigma-Aldrich. All of the purchased reagents were of analytical
grade and used as received.
