To achieve conditional deletion of SMAD4 specifically in CM, SMAD4fl/fl mice (Stock No. 017462, The Jackson Laboratory, Bar Harbor, Maine) were crossed with mice carrying the MerCreMer (MCM) transgene driven by the alpha-myosin heavy chain promoter (Myh6-Cre+/+) (Stock No. 005657, The Jackson Laboratory). Conditional Gene Knockout in Mice

Csf1rCre mice (#021024) and Smad4fl/fl mice (#017462) were purchased from Jackson Laboratory. The Smad2fl/fl mice were kind gifts from Dr. Daniel Bernard at McGill University. Human LangerinCre mice (9 (link)) were reported previously. All mice were backcrossed to C57BL/6J genetic background for at least six generations. The experiments were conducted in 7- to 12-week-old mice (both sexes) unless otherwise indicated. The mice were bred and maintained in a pathogen-free facility at the Laboratory Animal Services Center of Henry Ford Health System. Handling of mice and experimental protocols were in accordance with the Institutional Animal Care and Use Committee of Henry Ford Health System.
