LDH Activity Measurement in Oxidative Stress

The LDH activity of the cell supernatant in the Se cytotoxicity assay and oxidative stress model was determined using an LDH Cytotoxicity kit (Jiancheng Bioengineering Institute, Nanjing, China). Briefly, after exposure to 1000 μmol/L H2O2 and with/without 100 ng/mL Se for 24 h, a combination of 120 μL cell-free supernatant and 60 μL LDH reaction mixture was incubated at room temperature in the dark for 30 min. The cell-free supernatant of cells without the H2O2 and Se treatments served as the negative control. The absorbance was detected at a wavelength of 490 nm.
