Nitric Oxide Quantification in BALF

Eighty μL aliquots of BALF samples and standards (0–100 μM sodium nitrite (Sigma-Aldrich Co., St. Louis, MO, USA) dissolved in double distilled water) were pipetted into the 96-microplate wells (Nunc, Thermo Fisher Scientific, Rockford, IL, USA). One hundred sixty μL aliquots of Griess reagent were then added into each well to develop the color. The Griess reagent was freshly prepared from Reagents A and B at a ratio of 1 : 1 (Reagent A: 1% (w/v) sulfanilamide (Sigma-Aldrich Co., St. Louis, MO, USA) dissolved in 5.0% (v/v) phosphoric acid (Riedel-de Haen, Seelze, Germany); Reagent B: 0.1% (w/v) N-(1-naphthyl)ethylenediamine dihydrochloride (Sigma-Aldrich Co., St. Louis, MO, USA) dissolved in deionized water). After incubation for 5 min, the plate was read on a plate reader (ELISA reader, ASYS Hitech, GmbH, Austria) at 550 nm. Using the standard curve, the NO concentration for each unknown sample was determined [20 (link)].

Analytical Reagent-Grade Solvents Protocol

All solvents and reagents were of analytical reagent grade. Methanol and acetonitrile (HPLC grade) were purchased from Merck, Darmstadt, Germany. Acetate buffer was prepared by blending suitable amounts of acetic acid (0.2 M) with sodium acetate (0.2 M) in pH range of 3.6–5.6. Borate buffer (pH 5.5–13) was prepared by blending suitable amounts of boric acid (0.02 M) with sodium hydroxide (0.2 M). Dipotassium hydrogen phosphate was purchased from Sigma-Aldrich, Seelze, Germany. Phosphoric acid was from Riedel-deHaën (Seelze, Germany).

Hop Variety Characterization Protocol

Methanol (HPLC grade) was purchased from J. T. Baker (Deventer, The Netherlands); toluene, diethyl ether, phosphoric acid (85%) and hydrochloric acid (37%) were obtained from Honeywell (Charlotte, NC, USA); sodium hydroxide was purchased from Sigma Aldrich (St. Louis, MO, USA); international calibration extract 4 (ICE4) was obtained from Labor Veritas (Zürich, Switzerland). Hop material was supplied by Hmezad exim d.d., except for the Bobek cones, which were supplied by Farm Bosnar and the Styrian Wolf cones, which were supplied by Farm Bizjak, both located in the Savinja valley, Slovenia. Four different hop varieties (Celeia, Aurora, Bobek, and Styrian Wolf) were used in the experiment in the form of both cones and pellets, whereas Styrian Gold was used only in cone form and Savinjski Golding was used only as pellets. All used varieties were grown in Slovenia.

Preparation and Characterization of Antibiotic Solutions

Quantification of Polyphenolic Compounds

Determination of NSAIDs in Serum

Ketoprofen (≥98.0%, KET), naproxen (≥98.0%, NAP), sodium diclofenac (≥98.5%, DCF), and indomethacin (≥98.5%, IND) were purchased from Sigma-Aldrich (Darmstadt, Germany) and used as model analytes. The composite poly-ε-caprolactone sorbent was fabricated at the Technical University of Liberec from poly-ε-caprolactone (Mw 43.000) provided by Polysciences (Heddesheim, Germany), and chloroform (≥97%) and ethanol (99.97%) bought from Penta Chemicals (Pardubice, Czech Republic).

Coffee Husk Extraction and Analysis

Ethanol, sulfuric acid, and phosphoric acid, all of which were of analytical grade from Honeywell (Seelze, Germany), were used in the process. Sodium metabisulfite, used to stabilize the chlorogenic acids during the assays, was of analytical grade and was purchased from Sigma Aldrich (Steinheim, Germany). Chlorogenic acid was purchased from Biopurify (Chengdu, China). Trigonelline, sodium 3-(trimethylsilyl)-[2,2,3,3-d4]-1-propionate (TSP), deuterium oxide, and other chemicals used were purchased from Merck Millipore (Burlington, Massachusetts, USA). Coffee husks from the wet depulping and demucilaging process of coffee beans (Coffea arabica) were cleaned until water was clear. Raw coffee husk was split into lots of 3 kg and kept in a freezer at −20 degrees Celsius prior to experiments; batches of 1 kg of coffee husk were thawed at room temperature when needed.
