Pomegranate Antimicrobial Potential Analysis

Zn (II), as zinc sulfate heptahydrate (ZnSO4·7H2O), and potassium hydrogen phthalate were obtained from ThermoFisher Scientific (Loughborough, UK). Punicalagin (≥98%), Folin–Ciocalteu (F–C) reagent, ascorbic acid, and sodium carbonate (Na2CO3) were all obtained from Sigma-Aldrich (Gillingham, UK). Mueller–Hinton broth (MH broth), Mueller–Hinton agar (MH agar), and Brain–Heart Infusion agar (BHIA) were obtained from Oxoid (Basingstoke, UK). Phytochemical Extraction and Bioassays

Absolute ethanol (EtOH) for the extraction of plant samples was purchased from HmBg Chemicals Inc. (Germany). For the α-glucosidase inhibitory assay, quercetin, glycine, 4-nitrophenyl-α-D-glucopyranoside and α-glucosidase from Saccharomyces cerevisiae were purchased from Sigma Aldrich (St. Louis, USA). For liquid chromatography mass spectrometry (LC-MS) analysis, HPLC grade methanol, acetonitrile, acetic acid and hydrochloric acid were purchased from Merck (Darmstadt, Germany). Liquid nitrogen was supplied by MOX Company (Petaling Jaya, Malaysia).

Synthesis and Characterization of Stimuli-Responsive Hydrogels

Optimized Cell Culture Conditions for Diverse Cell Lines

All chemicals were of analytical grade and used as received without further purification. Deionized water was used for all experiments. All experiments were conducted under ambient atmosphere. Calcium chloride (CaCl2), ethyl alcohol, sodium carbonate (Na2CO3), polyethyleneimine (PEI, Mw =70 000) and arginine were purchased from Sigma-Aldrich (St Louis, MO). Plasmids were stored at −20°C. The rabbit anti-GAPDH antibody was purchased from Cell Signaling Technology. P53 antibody (Abcam, USA) reactions were visualized with an anti-rabbit IR secondary antibody (LICOR, USA).
Human cell lines, Hep3B, QSG-7701, H1299, 293a and Hela cells were obtained from American Type Culture Collection (Rockville, MD). All cells were maintained in Dulbecco’s modified Eagle’s medium (DMEM, Life Technologies) with 10% fetal bovine serum (FBS, Gibco) at 37°C in a 5% CO2 humidified incubator.

Chitosan-based Hydrogel Fabrication

Silk Fibroin Solution Preparation Protocol

Silk fibroin solutions were prepared following an established protocol [54 (link)]. Whole cocoons (OliverTwistsFibres, Durham, UK) were cut into small pieces and were boiled in a 0.02 M aqueous solution of sodium carbonate (Na2CO3) (Sigma-Aldrich, St. Louis, MO, USA) for 30 min to degum the fibroin fibers. The remaining fibers were rinsed and allowed to dry overnight in ambient conditions. The dry fibroin was then dissolved in a 9.3 M aqueous solution of Lithium Bromide (LiBr) (Sigma-Aldrich, St. Louis, MO, USA) at 60 °C for 4 h. This solution was placed into a set of dialysis cassettes (Thermo Fisher Scientific, Waltham, MA, USA) and spun in milli-Q water for 48 h. The milli-Q water was changed a total of 6 times over the 48 h. The remaining solution was removed from the cassettes and centrifuged at 4 °C and 4800 rpm for 20 min. This was repeated to ensure purity.
