Luciferase Reporter Assay for Gene Regulation

Check if the same lab product or an alternative is used in the 5 most similar protocols
pGL3-GPT2 promoter-LUC was co-transfect with an internal control Renilla luciferase expressing CMV-Renilla. Forty-eight hours after transfection, cells were collected for Luciferase assay according to the manufacturer's instructions (Promega). Luminescence was measured with a Lumat LB Single Tube Luminometer (Berthold). Relative luciferase activity was expressed as the ratio of luminescence normalized to the internal control. The luciferase reporter assay was performed in triplicate in at least three separate transfection experiments.

Dio2 Luciferase Assay in SCC Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
The reporter plasmids (Dio2-LUC-WT, Dio2-LUC-Δ1, Dio2-LUC-Δ2, Dio2-LUC-Δ3, Dio2-LUC-mut, TRE3-TK-LUC) and CMV-Renilla were co-transfected into SCC cells. LUC activities were measured 48 h after transfection with the Dual-Luciferase Reporter Assay System (Promega, Madison, Wisconsin, USA, cod. E1910). Luminescence was measured with a Lumat LB Single Tube Luminometer (Berthold). Differences in transfection efficiency were corrected relative to the level of Renilla Luciferase. Each construct was studied in triplicate in at least three separate transfection experiments.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
