Lichenase-treated extracts of S. viridis vegetative tissues and grain samples were purified using solid phase extraction (SPE) cartridges packed with graphitized carbon (Varian Bond Elut Carbon 50 mg ml−1 columns) according to Ermawar et al. The ratio of DP3:DP4 oligosaccharides in the vegetative tissue extracts were analysed using either high-performance liquid chromatography (HPLC) [91 (link)] or HPAEC-PAD on a Dionex ICS-5000 chromatograph. Analyzing Starch and β-Glucan in Flour Samples

A small-scale version of the Megazyme Total Starch Assay (20 mg of samples), which is based on the amyloglucosidase⁄α-amylase method (McCleary 1994), was developed to estimate starch content in the flour samples. Analyses of (1,3;1,4)-β-glucan in wheat endosperm and bran were performed using a scaled down Mixed-Linkage β-Glucan Assay Kit (Megazyme International Ireland Ltd, Wicklow, Ireland), which is based on the industry standard method for barley (22) , with 15 mg of samples. To determine the ratio of the degree of polymerization (DP) of major oligosaccharide components of the (1,3;1,4)-β-glucan, which are expressed as a DP3:DP4 ratio, the oligosaccharides released by lichenase digestion during the Megazyme assay were subjected to solid phase extraction on Varian Bond Elut Carbon 50 mg/1ml columns, eluted with 55% acetonitrile, before they were analysed with the HPAEC-PAD (Dionex, model ICS-5000 LC).
