Nitazoxanide Quantification in Pharmaceuticals

Nitazoxanide (NTX) was supplied by Alandalous medical company, Egypt. Its purity was certified to be (99.7 ± 0.72%). Anhydrous citric acid (99.9%) and l-cysteine (98%) were purchased from Loba—Chemie (Mumbai, India). Analytical reagent grade of boric acid, disodium hydrogen phosphate, sodium hydroxide, phosphoric acid, sulphuric acid, hydrochloric acid, methanol, ethanol, acetone, isopropyl alcohol and acetonitrile were obtained from El-Nasr chemical company, Cairo, Egypt. Surface active agents, cetrimide, Tween 80 and sodium dodecyl sulfate were supplied from Sigma Aldrich, Germany. Plasma samples were kindly supplied from El-Shatby University Hospital, Blood Bank, Alexandria, Egypt, and were stored frozen until use. Fresh deionized water was used throughout the work. Pharmaceutical preparations involved in this study are Nanazoxid® oral suspension labeled to contain 100 mg NTX/5 mL and Nanazoxid® film coated tablets labelled to contain 500 mg NTX (Utopia Company for Pharmaceuticals Industry, Cairo, Egypt).

Analytical Characterization of NFX Drug Compounds

NFX (purity% = 99.8 ± 0.82%) was kindly provided by Amoun Pharmaceutical Company, Egypt. The whole study was conducted using analytical-grade reagents. Disodium hydrogen phosphate, boric acid, sodium hydroxide, phosphoric acid, hydrochloric acid, sulphuric acid, dimethyl sulphoxide, methanol, acetone, ethanol, isopropyl alcohol and acetonitrile were purchased from El-Nasr Chemical Company, Cairo, Egypt. Anhydrous citric acid (purity% = 99.9%), and l-cysteine (purity% = 98%) were purchased from Loba Chemie (Mumbai, India). Surface active agents including Tween, cetrimide, and sodium dodecyl sulfate were brought from Sigma-Aldrich, Germany. Commercial pharmaceutical dosage forms, namely, Antinal® oral suspension (labeled to contain 220 mg NFX/5 mL) and Antinal® capsules (labelled to contain 200 mg NFX), (products of Amoun Pharmaceutical Company, Cairo, Egypt), were bought from a local pharmacy. Fresh deionized water was utilized as the diluting solvent throughout the entire work.

Synthesis and Biological Evaluation of Methyl Cinnamate Derivatives

Metformin hydrochloride and methyl cinnamate were purchased from Fluorochem (Hadfield, Derbyshire, UK); dicyandiamide, aniline, diethyl oxalate, methyl salicylate and sodium methoxide were purchased from TCI Chemical (Tokyo, Japan); isopropyl palmitate was purchased from Sigma Aldrich (St. Louis, MO, USA); methyl benzoate was gifted by the Department of Chemistry, Chulalongkorn University. Calcium chloride dihydrate and trisodium citrate dihydrate were purchased from Merck (Darmstadt, Germany). Citric acid anhydrous was purchased from Loba Chemie PVT. LTD (Mumbai, India). All of solvents used were distilled or analytical grade.
HCT116 (Human colorectal carcinoma) and SW620 (Human colorectal adenocarcinoma) cells were obtained from Nanomedicine RU, Faculty of Medicine, Chulalongkorn University. DMEM (Dulbecco’s Modified Eagle Medium), RPMI, fetal bovine serum (FBS), antibiotics, trypsin-EDTA, PBS (Phosphate Buffered Saline) (1X, pH 7.4) and Prestoblue™ cell viability reagent were purchased from Thermo Fisher Scientific (Waltham, MA, USA). Cisplatin was purchased from Sigma Aldrich (St. Louis, MO, USA) and dialysis bag Spectra/Por membranes (MWCO = 12–14 kDa) were purchased from Spectrum Laboratories, Inc. (Rancho Dominguez, CA, USA).

Quantitative Hg2+ Analysis in Skincare

All the
experiments were performed in aqueous solution. Citric acid anhydrous
99.5% (Loba Chemie Pvt., Ltd., India) and urea 99.5% (Loba Chemie
Pvt., Ltd., India) were used for CD synthesis. The standard solution
of 1000 mg/L Hg2+ was prepared by dissolving mercury(II)
acetate (Sigma-Aldrich, USA) in deionized water and used as a stock
solution. The working standard solution of 0.3–600 mg L–1 Hg2+ was prepared by diluting the stock
solution. Acetate buffer (0.10 M) at pH 6 and 7 was prepared from
acetic acid (Carlo Erba, Italy) and sodium acetate (Sigma-Aldrich,
USA). Phosphate buffer (0.1 M) at pH 6 was prepared from sodium phosphate
(Sigma-Aldrich, USA). All of the solutions were prepared in deionized
water.
In this work, we aimed to quantitatively analyze Hg2+ in skincare products. Different products for skin conditions,
particularly whitening cream, were purchased from the online market,
local cosmetic shops, and supermarkets in Nakhon Pathom, Thailand.
For analysis, the exact weight of 0.10 g of the sample was dissolved
in 0.50 mL of 5% (v/v) HNO3 (Carlo Erba, Italy), and the
volume was made to 25 mL in a volumetric flask by 0.1 M acetated buffer
at pH 7.0, which was then injected to the SIA system. All samples
were analyzed using the proposed flow system and the ICP-MS reference
method.
