Measuring Oxidative Stress in MEFs

Quantifying Oxidative Stress and CD73 Effects

Cells (106/mL) were incubated in serum‐free RPMI 1640 medium containing 2 μmol/L 5‐(and‐6)‐chloromethyl‐2′,7′‐dichlorodihydrofluorescein diacetate in the absence or presence of 10 ng/mL lipopolysaccharide at 37°C for 30 minutes, washed with PBS, and analyzed using flow cytometry. To determine effect of CD73 on ROS, 100 μmol/L AMP and 100 μmol/L APCP were added 30 minutes before lipopolysaccharide.
