Giemsa Staining for Macrophage Infection

Check if the same lab product or an alternative is used in the 5 most similar protocols
After 4 and 24 h of infection, the chamber slides were washed two times with PBS 1× and were then fixed with acetone/methanol (1:1 v/v; Sigma-Aldrich, St. Louis, MO, USA) and were incubated at −20 °C for 24 h followed by Giemsa staining (Fast Panotic Kit, Laborclin, São Paulo, Brazil). At least 600 macrophages were counted per well to achieve the parameters of the Macrophage Infection Rate [(infected macrophage/total macrophage) ∗ 100], amastigotes per infected macrophage, and the infection index, which was calculated by multiplying the rate of infected macrophages by the average number of amastigotes per macrophage.

Leukocyte Count in Temporomandibular Joint

Check if the same lab product or an alternative is used in the 5 most similar protocols
To analyze the total leukocyte count, 10 μl of EDTA (1 mM) were used to wash the articular cavity of the temporomandibular joint. Afterwards, 90 μl of PBS was added and the exudate was dilute in Türkś stain (1:2) and leukocytes were analyzed in a Neubauer chamber in an optical microscope (100 x). The results are expressed as cells x 10 4 per joint. For differential count (neutrophils and lymphocytes), joint exudate was submitted to cytocentrifugation and stained by fast Panotic kit (LaborClin, Pinhais, PR, Brazil) and were analyzed in an optical microscope (1000 x). The results are expressed as percentage of each cell type/total leukocyte counts x 10 4 per cavity.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
