Dietary Peptides Modulate Colonic Inflammation

IRW and IQW are synthesized by Ontores (Zhejiang Province, China). The purity of the two small peptides is 99%. Thirty-two 8-week-old mice weighing on average 23 g were randomly assigned to four groups of 8 each: basal diet [17 (link), 18 (link)] (CON), basal diet with 5% DSS (Meilun Biotechnology, Dalian Province, China, M.W: 5000), 0.03% IRW with 5% DSS (IRW-DSS), and 0.03% IRW with 5% DSS (IQW-DSS). The animals were housed with a 12-h light/dark rotation (the lighting time is 8 a.m.); the ambient temperature was controlled at 22–24°C and the humidity 55%–60%. After an adaptation period of 3 days, 5% DSS was diluted in water and all mice were fed the basal diet and drinking their respective water ad libitum; daily weight changes of mice were recorded. DSS was used for 7 days in mice, and on day 8, all mice were slaughtered. Serum samples were collected to determine the level of amino acids; colonic tissue was quick-frozen for the determination of gene expression. This study was carried out by the Chinese Guidelines for Animal Welfare and approved by the Animal Care and Use Committee of the Institute of Subtropical Agriculture, Chinese Academy of Sciences (0207).

DSS-Induced Colitis in Balb/c Mice

A total of 27 specific pathogen-free male Balb/c mice (6 weeks old, from Jinan Peng Yue Experimental Animal Technology Co., Ltd.), weighing 18±2 g, were kept in the Animal Laboratory of Jining Medical University. The animal experiments were approved by the Animal Ethics Committee of Jining Medical University (approval no. JNMC-2020-DW-ZXY-001). If weight loss reached 15-20% of the original weight within 72 h, then the mice were sacrificed by cervical dislocation. After 3 days of adaptive feeding, mice were randomly divided into the following three groups (n=9 per group): Control group, model group, and treatment group. For DSS-induced colitis, 5% (w/v) DSS (Meilun Biotech Co., Ltd.) was added to the drinking water in the model group and treatment group for 7 days. After the establishment of the DSS-colitis model, mice in the control and model groups were administered normal saline, while mice in the treatment group were administered PF (20 mg/kg/day). All administration methods were by gavage, and the mice were dosed continuously for 7 days. Mice from each group were sacrificed by cervical dislocation after 14 days.

Exploring Dextran Sulfate Sodium-Induced Colitis in Mice

We purchased 18 C57BL/6 male mice (age: 6–8 weeks) from Jiangsu Jicuiyaokang Biotechnology Co (Nanjing, China). The mice were housed at the Jiangxi Academy of Sciences, Royo Biotech Co., Ltd. (Nanchang, China). They were housed at a temperature of 25°C with a 24-h light-dark cycle. Mice had ad libitum access to food and water. After 7 days of acclimation, the mice were divided into three groups (n=6 per group): (1) Control group that was allowed ad libitum access to food and water; (2) DSS group: ad libitum diet was allowed for 1–7 days and 3% DSS (Meilunbio, Dalian) was added to the drinking water for 8–14 days; (3) Arec group: ad libitum diet was allowed for 1–7 days, 3% DSS was added to the drinking water for 8–14 days, and 5 mg/kg arecoline (Yuanye, Shanghai) was administered daily by tube feeding.

Institute (Nanjing, China). Mouse IL-6, TNF-α ELISA kits, and
Goat anti-rabbit IgG were purchased from Boster Biological Technology
Co. Ltd. (Wuhan, China). The bicinchoninic acid (BCA) protein assay
kit was purchased from Solarbio Science & Technology Co., Ltd.
(Beijing, China). GADPH antibody was obtained from Abcam (Cambridge,
MA). The occludin antibody was purchased from Proteintech (Wuhan,
China). Fixable Viability Stain 780 from BD Pharmingen is useful for
discrimination of viable from nonviable cells. Antibodies for CD45,
CD3, CD4, CD11b, and CD16/CD32 (Fc Blocker) were bought from BD Pharmingen.
Antibody for F4/80 was from Biolegend. RPMI 1640 and fetal bovine
serum (FBS) were purchased from Gibco. Penicillin, streptomycin, and
glutamine were from Solarbio Life Technologies. Collagenase VIII and
DNAse I were from Sigma-Aldrich. Other chemicals were of analytical
grade.

Induction and Analysis of Murine Colitis

Six-to-eight-week-old female C57BL/6 (wild-type, WT) mice were obtained from the Hunan Silaike Jingda Laboratory Animal Co., Ltd., (Changsha, China), and miR-10b knockout (KO) mice (generated on C57BL/6 background) were purchased from Nanjing Biomedical Research Institute of Nanjing University (Nanjing, China). All mice were fed autoclaved food and water and maintained in a specific-pathogen-free facility at Hunan Normal University (Changsha, China). To induce colitis, WT and miR-10b KO mice were given drinking water containing 4% DSS (MW 36,000–50,000; Meilunbio, Dalian, China) for 7 consecutive days. Mice in control groups were given normal drinking water only. Survival rates, body weight loss, rectal bleeding, and stool consistency were monitored daily. The disease activity index (DAI) scores were the sum of body weight loss, fecal hardness, and bleeding scores as previously described (Supplementary Table S1) [16 (link)]. After 7 days of DSS treatment, mice were euthanized by CO2 inhalation. Serum and colon samples were harvested and stored in a refrigerator at −80 °C for subsequent experiments.

Histological Evaluation of DSS-Induced Colitis

Dose-dependent effects of DSS on NIH3T3 cells

DSS (purity >98%; molecular weight 198.17), purchased from Meilunbio (Dalian, China), was dissolved in sterile distilled water as a 40-mM stock solution. NIH3T3 cells (Procell Life Science & Technology Co., Ltd., Wuhan, China) were cultured in Dulbecco’s modified eagle medium (DMEM, Gbico, USA) containing 10% fetal bovine serum (FBS) in a 37°C, 5% CO2 incubator. Then, NIH3T3 cells were treated in different ways as followed: part of NIH3T3 cells were treated with DSS at different concentrations (0, 25, 50, 100, 200, and 400 μM) for 24 h; part of NIH3T3 cells were pretreated with various concentrations of DSS (0, 25, 50, 100, 200, and 400 μM) for 2 h, followed by stimulation with or without TGF-β1 (5 ng/mL) for 24 h and another part of NIH3T3 cells were pretreated with DSS (50 μM) for 2 h, followed by the stimulation with or without TGF-β1 (5 ng/mL) for 24 h.
