Quantifying Islet Cell Populations

Characterizing CAR T Cell Phenotype

To stain for CAR T cells we used anti-CD3 clone 7D-6 (Thermo-Fisher; Waltham, MA), anti-CD8 clone 3B5 (Thermo-Fisher; Waltham, MA), and cetuximab (Eli Lilly, Indianapolis, IN) conjugated with a lightning link labeling kit (Innova Biosciences, Cambridge, UK). Viability was ascertained with 7AAD exclusion (Thermo-Fisher; Waltham, MA). Flow cytometry was performed on a BD LSR II (BD biosciences; San Jose, CA), and analyzed with FlowJo (FlowJo LLC; Ashland, Oregon).
