Multicolor Flow Cytometry of Pancreatic Islet Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
Flow cytometry of pancreatic islet cells was performed as previously
described60 (link). Islet
cells were fixed and permeabilized with the Foxp3 staining kit (eBioscience) and
then stained with APC-conjugated antibody to insulin (clone 182410, R&D
Systems), Alexa Fluor 488–conjugated antibody to GFP (Invitrogen) and
antibody to glucagon (clone 199017, R&D Systems; PerCP-Cy5.5 labeled
using the Lightning-Link labeling kit from Innova Biosciences).

Quantifying Islet Cell Populations

Check if the same lab product or an alternative is used in the 5 most similar protocols
Flow cytometry was performed on pancreatic islet cells fixed and permeabilized with the Foxp3 staining kit (eBioscience) and stained with anti-insulin APC (clone 182410, R&D Systems) and anti-glucagon PerCP-Cy5.5 (clone 199017, R&D Systems) labeled using the Lightning-Link labeling kit (Innova Biosciences). Data were acquired on a FACSCanto II (BD) and analyzed with FlowJo software (Tree Star).

Characterizing CAR T Cell Phenotype

Check if the same lab product or an alternative is used in the 5 most similar protocols
To stain for CAR T cells we used anti-CD3 clone 7D-6 (Thermo-Fisher; Waltham, MA), anti-CD8 clone 3B5 (Thermo-Fisher; Waltham, MA), and cetuximab (Eli Lilly, Indianapolis, IN) conjugated with a lightning link labeling kit (Innova Biosciences, Cambridge, UK). Viability was ascertained with 7AAD exclusion (Thermo-Fisher; Waltham, MA). Flow cytometry was performed on a BD LSR II (BD biosciences; San Jose, CA), and analyzed with FlowJo (FlowJo LLC; Ashland, Oregon).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
