PSMA-Targeted Paclitaxel-Loaded Nanomedicine

PLGA–COOH (50: 50, MW = 12.3 kDa) was purchased from Jinan Daigang Biomaterial Co. (Shandong, China). Paclitaxel was purchased from Meilun Bio (Dalian, China). Perfluoropentane (PFP), polyvinyl alcohol (PVA), IR780, N-(3-dimethyl-aminopropyl)-N′-ethylcarbodiimide hydrochloride (EDC), 2-(N-morpholino) ethanesulfonic acid (MES monohydrate) and 1,1′-dioctadecyl-3,3,3′,3′ tetramethylindocarbocyanine perchlorate (DiI), and 4′,6-diamidino-2-phenylindole (DAPI) were obtained from Sigma-Aldrich (St. Louis, MO, USA). PSMA monoclonal antibody was purchased from Cell Signaling Technology (USA). Goat anti-human IgG Fc (FITC) was purchased from Abcam (Shanghai, China). Cell Counting Kit-8 (CCK-8) was obtained from Dojindo Molecular Technology (Shanghai, China). RPMI-1640 mediumand fetal bovine serum (FBS) were purchased from Gibco (USA). All other reagents used in this work were of analytical grade and were used as received.

Preparation of PTX-loaded PLGA Nanobubbles

PTX-loaded PLGA NBs (PTX-PLGA NBs) were generated using a water/oil/water double emulsion. Briefly, 50 mg PLGA-COOH (50:50, molecular weight [MW] =12.3 kDa, Jinan Daigang Biomaterial Co., Ltd, Shandong, People’s Republic of China) was directly dissolved in 1.5 mL of dichloromethane and mixed with 5 mg of PTX (Haoxuan, Xian, People’s Republic of China) and 0.2 mL of Milli-Q water under sonication (VCY-500; Yongyan, Shanghai, People’s Republic of China) with an amplitude of 20% for 1.5 min to obtain the first emulsion (first emulsion). The first emulsion was immediately poured into 8 mL of a poly(vinyl alcohol) (PVA; Sigma-Aldrich Co., St Louis, MO, USA) solution (4%, w/v) and sonicated for 1 min to form a double emulsion (second emulsion). Following dichloromethane evaporation to solidify the PTX-PLGA, the double emulsion was transferred into 20 mL of isopropanol (2%, v/v) and continuously stirred for 2–4 h at room temperature to extract dichloromethane. The obtained PTX-PLGA nanoparticles were further collected through centrifugation at 8,000 rpm for 10 min and washed three times using Milli-Q water, subsequently lyophilized for 48 h, and filled with perfluoropropane gas. A lyophilized powder of PTX-PLGA NBs was obtained and sealed at 4°C until further use. NBs without the addition of PTX were used as pure PLGA NBs.

Perfluoropentane-encapsulated PLGA Nanoparticles

PCM (WLSEAGPVVTVRALRGTGSW) and FITC-PCM were purchased from GL Biochem Ltd. (Shanghai). PLGA-COOH (50:50, MW ¼ 20,000) was obtained from Jinan Daigang Biomaterial Co., Ltd. (China). Perfluoropentane (PFP), MES hydrate, 1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide hydrochloride (EDC), and N-hydroxysuccinimide (NHS) fluorescent dyes, including 4′,6-diamidino-2-phenylindole (DAPI) and 1,1′-dioctadecyl-3,3,3′,3′-tetramethylindocarbocyanine perchlorate (DiI), were obtained from Sigma-Aldrich Chemical Co. (St. Louis, MO, USA). 17β-estradiol (E2) was purchased from Solarbio (China). Deionized water was used in all experiments.

Synthesis and Characterization of Oleic Acid-Coated SPIONs

Oleic acid-coated SPIONs (d = 10 nm, 5 mg/mL) were purchased from Xi’an ruixi Biological Technology Co., Ltd. (China). CHP and fluorescein isothiocyanate (FITC)-CHP were purchased from GL Biochem Ltd. (Shanghai, China). PLGA-COOH (50:50, MW ¼ 20,000) was obtained from Jinan Daigang Biomaterial Co., Ltd. (China). Melatonin was purchased from Sigma-Aldrich (St. Louis, MO, USA). MES (2-(N-morpholino)ethane sulfonic acid) hydrate, EDC (1-ethyl-3-(3-dimethylaminopropyl)-carbodiimide hydrochloride), NHS (N-hydroxysuccinimide) were obtained from Sigma-Aldrich Chemical Co. (St. Louis, MO, USA). The fluorescent dyes DAPI (4′,6-diamidino-2-phenylindole), DiI (1,1′-dioctadecyl-3,3,3′,3′ tetramethylindocarbocyanine perchlorate) and DiR (1,1ʹ-dioctadecyl-3,3,3′,3′-tetramethylindotricarbocyanine iodide) were also obtained from Sigma-Aldrich Chemical Co. Deionized water was used in the preparations.
