Check if the same lab product or an alternative is used in the 5 most similar protocols
Anti-FLAG (M2) Cy3-labeled antibody was from Sigma (St. Louis MO). Anti-SAP97 antibody from Stressgen (Enzo Life Sciences, Farmingdale, NY), anti-SNX27 (ab77799) from Abcam (Cambridge MA). Anti β-actin antibody (sc-47778) from Santa Cruz Biotechnology (Santa Cruz, CA). Myristoylated PKA inhibitory peptide, 14–22 amide myristoylated Myr-N-Gly-Arg-Thr-Gly-Arg-Arg-Asn-Ala-Ile-NH2 (PKI) was purchased from Calbiochem (EMD-Millipore, Billerica MA). Isoproterenol, alprenolol, dithiothreitol, poly-L-lysine (P0899), leupeptin, aprotinin, chymostatin and phenyl methyl sulfonyl fluoride were from Sigma Chemical Co. (St. Louis, MO). Flexi siRNA oligo sequence to human SNX27 5′-AACCAGGTAATTGCATTTGAA-3′ and its inactive control 5′-CCATAATACAAG GTATAA-3′ were purchased from Qiagen (Valencia CA). Human embryonic kidney-293 (HEK-293) cells (CRL-1573) were from ATCC (Manassas, VA). HEK-293 cells passages 40–46 were grown in DMEM supplemented with 10% fetal bovine serum.

HEK 293 and THP-1 Cell Culture Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
HEK 293 (CRL-1573™) cells were obtained from American Type Culture Collection (ATCC, Manassas, MDVA, USA) and cultured in Dulbecco’s modified Eagle’s medium (Gibco™, Waltham, MA, USA; 11965118) with 10% fetal bovine serum (Gibco™, 16000044) and 1% antibiotic-antimycotic (Gibco™, 15240062) in a humidified atmosphere in a 5% CO2 incubator at 37°C. THP-1 (TIB-202) cells were obtained from ATCC and maintained in RPMI-1640 Medium (Gibco™, 21875034) containing 10% fetal bovine serum, 1% antibiotic-antimycotic and 0.05 mM 2-mercaptoethanol (Gibco™, 21985023). THP-1 cells were then stimulated with 50 nM phorbol myristate acetate (Sigma-Aldrich, St. Louis, MO, USA; P8139) for 24 h for induction of differentiation into macrophages.

HEK293 Cell Cultivation for Reagent Purification

Check if the same lab product or an alternative is used in the 5 most similar protocols
HEK293 CRL-1573 cells were obtained from the American Type Culture Collection (Manassas, VA). The identity of these cells was not subsequently authenticated because the cells were only used for the purification of reagents rather than as experimental materials. The cells were grown in Dulbecco’s Modified Eagle Medium +10% Fetal Bovine Serum (Invitrogen, ThermoFisher Scientific) in the presence of 100 units/ml of penicillin and 100 μg/ml of streptomycin and a 1:100 dilution of the antimycotic Fungizone (Gibco, ThermoFisher Scientific).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
