Isolation of PBMC from Whole Blood

Peripheral blood mononuclear cells (PBMC) were enriched by centrifuging the tubes at 1,700xg for 20 minutes at 25°C. After removing a 1 mL aliquot of plasma, the mononuclear cells were poured into a 50 mL conical tube and washed twice with MACS buffer (PBS containing 0.5% fetal bovine serum and 2 mM EDTA). The washed cell pellet was then suspended in 50 μl MACS buffer containing human Fc block (BD #564219) prior to staining.
