Whole blood was collected via cardiac puncture using a 1 mL BD Slip-Tip Disposable Tuberculin Syringe (28-gauge; BD Biosciences) and mixed with a 0.5 M EDTA pH 8.0 solution (1:10 ratio; AmericanBio, Natick, MA, USA). CBC analysis, including the number of white blood cells, and percentage of lymphocyte, monocyte, neutrophil, eosinophil, and basophil was measured using a Hemavet 950FS (Drew Scientific, Dallas, TX, USA) according to the manufacturer’s instructions.

Murine Blood and Bone Marrow Analysis

Check if the same lab product or an alternative is used in the 5 most similar protocols
Peripheral blood was collected from mice via submandibular bleeding and blood counts were recorded using the Hemavet 950fs (Drew Scientific). Blood counts were tabulated graphically with error bars corresponding to standard error of the means and compared using 2-tailed ttests. Bone marrow cellularity was determined using hemocytometer under light microscope.
Total marrow was incubated with lineage markers (CD3, CD4, B220, Ter119, CD11b) and Sca1+/c-Kit+ cells in lineage negative cells determined via flow cytometry on a BD LSRII instrument (BD Biosciences). Data was analyzed on FlowJo software (Treestar). Values were tabulated graphically with error bars corresponding to standard error of the means and compared using 2-tailed t-tests.

White Blood Cell Quantification

Check if the same lab product or an alternative is used in the 5 most similar protocols
To measure the number of white blood cells in blood upon challenge, 200 μl of blood were collected into BD Microtainer® Blood Collection Tubes (BD Biosciences). Blood samples were analyzed immediately using an Hemavet 950 FS (Drew Scientific). The results were compared to a mouse blood standard (Drew Scientific).

Submandibular Blood Collection for Hematology

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was collected by submandibular vein puncture into Microtainer EDTA tubes (BD Biosciences) for hematologic analysis (Hemavet 950 FS; Drew Scientific, Inc., Dallas, TX) or Microtainer serum separator tubes (BD Biosciences) for serum chemistry analysis on a Liasys 330 AMS Diagnostic liquid chemistry analyzer (Weston, FL). Investigators were blinded to treatment groups during analysis.

Monitoring Animal Health in Research

Check if the same lab product or an alternative is used in the 5 most similar protocols
All live animals were monitored daily for clinical signs under anesthesia twice a day. Body temperature was measured by inserting a soft probe attached to an Omron electronic digital stick thermometer (MC-BOMR, Omron Co.) into the rectum 2 cm from the anal margin for one minute [18] (link). All clinical signs were monitored from day 2 p.i.. Venous blood of biological indicators such as blood cell count using the Veterinary Multi-species Hematology System (Hemavet 950FS, Drew Scientific Co.) and evaluation of the viral load were performed.

Postoperative Blood Analysis Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
At time points one and five weeks post-surgery, blood was collected into EDTA-coated tubes for complete blood count analysis (CBC) performed using a Hemavet 950FS (Drew Scientific Inc., Waterbury, CT). Plasma was prepared for thrombin clotting time (TT), thrombin activity assay, and plasma inflammatory biomarkers. The TT was determined by Hemoclot® thrombin inhibitor assay, according to the manufacturer’s protocol (ANIARA, Mason, OH). The diluted TT is particularly sensitive to the effects of dabigatran, and displays a linear dose-response over therapeutic concentrations; therefore, dabigatran plasma concentrationwas calculated from diluted TT (26 (link)). Plasma thrombin activity was detected using SensoLyte® 520 thrombin activity assay kit, according to the manufacturer’s protocol (AnaSpec, Fremont, CA). Plasma cytokines were quantified using a mouse 96-well multiplex assay kit (MPXMCYTO-70K; Millipore, Billerica, MA), according to the manufacturer’s protocol. Bead fluorescence was detected using a BioRad Bioplex 200 suspension array reader and analyzed using Bio-Plex Manager 4.0 software (BioRad Laboratories, Hercules, CA).

Porcine Hematological Response Assessment

The pig BW and feed intake were recorded at the starting day and end of each
phase. Average daily gain (ADG), average daily feed intake (ADFI) and gain to
feed ratio (G:F) were calculated. Blood samples were collected from the jugular
vein of 8 pigs per treatment using 8 mL EDTA tubes. The blood samples were
analyzed to measure number of white blood cells and proportions of their
differentiation using a multi-parameter automated hematology analyzer calibrated
for porcine blood (Hemavet 950FS, Drew Scientific, UK).

Blood Characterization in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Blood was obtained from euthanized mice by cardiac puncture using 0.2M EDTA as an anticoagulant. Complete blood cell counting was done on an aliquot using a Drew Scientific Hemavet 950FS (Miami Lakes, FL. USA). Plasma was isolated from the remaining blood and aliquots were used to measure insulin by ELISA (Millipore; Burlington, MA. USA), and total plasma cholesterol, HDL, low density lipoprotein (LDL), and triglycerides using an Axcel Clinical Chemistry Analyzer (West Caldwell, NJ. USA). HOMA IR - scores were calculated as described (23 (link)).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
