Targeted Phenolic and Sugar Analysis

Acetonitrile and formic acid (both MS grade), methanol (high-performance liquid chromatography, HPLC grade), sodium hydroxide, sodium acetate, and hydrochloric acid were purchased from Merck (Darmstadt, Germany). Ultra-pure water (Thermofisher TKA MicroPure water purification system, 0.055 µS/cm; Bremen Germany) was used to prepare standard solutions and blanks. The solid phase extraction (SPE) cartridges used to concentrate the samples were Strata C18–E (500 mg/3 mL) obtained from Phenomenex (Cluster d.o.o., Belgrade, Serbia). Phenolic standards (protocatechuic acid, p-hydroxybenzoic acid, vanillic acid, syringic acid, ellagic acid, chlorogenic acid, caffeic acid, p-coumaric acid, ferulic acid, sinapic acid, cinnamic acid, rutin, hyperoside, cynaroside, apiin, naringin, astragalin, catechin, luteolin, apigenin, naringenin, kaempferol, aesculin, phloridzin, coniferyl aldehyde, and aesculetin) were supplied by Sigma Aldrich (Steinheim, Germany).
Sugar standards (trehalose, arabinose, glucose, fructose, sucrose, isomaltotriose, turanose maltose, and maltotriose) were purchased from Tokyo Chemical Industry (TCI, Europe, Belgium). Standard of sorbitol was obtained from Sigma Aldrich (Steinheim, Germany).

Synthesis and Antimicrobial Evaluation

All the chemicals and reagents used in the experimental part of the study were of analytical grade. Nutrient agar, nutrient broth, sabouraud dextrose agar, and sabouraud broth were purchased from HiMedia Laboratories (Mumbai, India). Aesculin, 2,2-diphenyl-1-picrylhydrazyl (DPPH), anilines, hydrochloric acid were purchased from Sigma Aldrich (Germany), LobaChemie (Mumbai, India), and SRL (Mumbai, India). Reaction progress was checked by thin-layer chromatography (TLC) method. Standard streptomycin, ampicillin, ciprofloxacin, and fluconazole were obtained from Belco Pharma, Bahadurgarh (India). The standard microbial strains E. coli 45, S. aureus 3160, P. aeruginosa 1934, C. albicans 183 and A. niger 282 were obtained in lyophilized form from MTCC, Chandigarh (India). Melting point was recorded by the Sonar melting point apparatus. FTIR spectra were recorded on Perkin Elmer FTIR spectrophotometer, 1H NMR, and 13C NMR spectra were recorded on Bruker Avance II 400 NMR spectrometer. Mass spectra were recorded on Waters Micromass Q-ToF Micro instrument while the elemental analysis was done by Perkin Elmer 2400 elemental analyzer.

Recombinant DGAS Purification Protocol
