Adult male Sprague-Dawley rats (200 to 220 g on arrival, 280 to 300 g at the time of experiments) and adult male Swiss mice (18 to 20 g on arrival, 22 to 25 g at the time of experiments) were purchased from JANVIER LABS (France) for algesimetry tests and biodistribution, respectively. They were housed in a standard controlled environment (22° ± 1°C, 60% relative humidity, 12-hour light/12-hour dark cycle, lights on at 8:00 a.m.) with food and water available ad libitum, without any handling for at least 1 week before being used for experiments. In all cases, experiments were performed in conformity with the guidelines of the Committee for Research and Ethical Issues of the International Association for the Study of Pain (37 (link)) and approved by the Animal Care Committee of the University Paris-Sud in accordance with the principles of laboratory animal care and the European legislation 2010/63/EU. All efforts were made to reduce animal numbers and minimize their suffering, as defined in the specific agreement (registered under no. 7493-2016102520355414).

Acute DFP Intoxication in Mice

Adult male Swiss mice (8 weeks, Janvier Labs, Le Genest-Saint-Isle, France) were housed four per cage with a 12 h/12 h light/dark cycle and food and water ad libitum. A period of 5-7 days of acclimation to housing conditions was observed. All animals were 9 weeks old when exposed to DFP. Animal experiments were approved by the ethics committee according to applicable French legislations, in compliance with the "3Rs" policy of reduction, re nement and replacement of animal use for scienti c procedures (Directive 2010/63/UE, décret 2013 - 118, approval number 9642-2017040316305252). Drugs DFP (Sigma Aldrich, L'Isle d'Abeau Chesnes, France) was freshly prepared by diluting the initial concentrated solution (0,1 g/ml in isopropanol) in ice-cold 0.9% (w/v) saline. The oxime HI-6 (1-2-hydroxyiminomethyl-1-pyridino-2-oxanopropane) dichloride was a generous gift of DRDC Su eld (Canada).
Atropine sulfate (AS, Sigma Aldrich, L'Isle d'Abeau Chesnes, France) and HI-6 were diluted in 0.9% (w/v) saline. Veterinary Diazepam (TVM, Lempdes, France) was used undiluted.
