B6 and Balb splenocytes were cultured in the presence and absence of syngeneic B6 membrane cells at 37°C in 5% CO₂. After 96 hours, culture supernatants were assayed for targets: IFNγ, IL-12p70, IL-13, IL-1β, IL-2, IL-4, IL-5, IL-6, TNFα, GM-CSF, IL-18, IL-10, IL-17A, IL-22, IL-23, IL-27, and IL-9 cytokines following the manufacturer’s (Thermo Fisher Scientific) protocol. Multiplex Cytokine Profiling of Mice

Cytokines were measured using a Bio-Plex 200 system (Bio-Rad, Hercules, CA) and BioManager software (Bio-Rad, Hercules, CA) for analysis. The following multiplex kits were purchased from Merck Millipore (Billerica, MA): (i) the MCYTOMAG-70K Mouse Cytokine/Chemokine kit, containing IL-1α, IL-1β, IL-6, IL-7, IL-10, IL-12p40, IL-12p70, IL-15, macrophage inflammatory protein 1α (MIP-1α), MIP-1β, MIP-2, RANTES and tumor necrosis factor alpha (TNF-α); (ii) the MCYP3MAG-74K Mouse Cytokine/Chemokine kit, containing IL-23 and IL-27. The kits were run according to the manufacturer’s instructions, with the exception of sample collection and processing as described in the previous subsection. The samples were incubated for 2 hours at room temperature (25°C). All the samples were run on a single plate so as to avoid plate to plate variability.

Cytokine Profiling of Splenocyte Cultures

Freshly isolated splenocytes were cultured either alone or with mitomycin C treated C57Bl/6J splenocytes (donor-type stimulator cells). After 48 hours, culture supernatants were collected and frozen. Samples were later thawed and assayed using the Milliplex Map Mouse Cytokine/Chemokine Premixed 25 Plex kit (Millipore, Billerica, MA). Cytokines were measured with the Luminex 100 platform (Luminex, Austin, TX) with BioManager Software (BioRad, Hercules, CA) used for analysis as previously described.^{24 (link)}
