Enzyme Immobilization Protocol

Isopropyl‐β‐D‐thiogalactopyranosid (IPTG) was obtained from Carl Roth GmbH and Co. KG. Protease inhibitor cocktail set VII from Calbiochem was purchased from Merck KGaA and PLP from Sigma‐Aldrich. Immobilization supports Lifetech™ ECR were kindly donated by SpinChem AB and Purolite®. Enzyme carrier Eupergit® CM was purchased from Sigma‐Aldrich. D‐asparagine monohydrate was purchased from Alfa Aesar and L‐asparagine monohydrate from Sigma Aldrich chemicals, both of purity ≥99%.

Immobilized Mandelate Racemase Protocol

The pET-15b plasmid with mandelate racemase gene was kindly donated by Prof.
Stephen L. Bearne, Dalhousie University, Halifax, Canada [15] . Supports used for enzyme immobilization were Eupergit ® CM (Sigma-Aldrich, St. Luis, MO, USA) and CNBr-activated Sepharose 4 Fast Flow (GE Healthcare Bio-Sciences AB, Uppsala, Sweden). Protease inhibitor cocktail set VII from Calbiochem was purchased from Merck, Darmstadt, Germany and Rotilabo PES filters with 0.2 µm pores from Carl Roth, Karlsruhe, Germany.
Two Chirobiotic T columns were used (Sigma Supelco, St. Louis, MO, USA). One (column C1) was applied in concentration analysis (5 µm, L=150 mm, d=4.6 mm) and another one (column C2) was connected with the outlet of the fixed-bed (16 µm columns, L=150 mm, d=10 mm). The mobile phase is discussed under 2.3 and 2.6.
Substrates used for enzymatic reactions were R-mandelic acid and R-3-chloromandelic aci d purchased f rom Si gma Aldrich Chemie (Sigma-Aldri ch, St. Luis, MO, USA), S-4methoxymandelic acid from Beta Pharma Scientific (Branford, CT, USA) and S-2chloromandelic acid from AK Scientific (Union City, CA, USA).
