Real-Time Cell Analysis of Invasion, Migration, and Proliferation

Cell invasion, migration and proliferation were evaluated using the Real-Time Cell Analyzer - Dual Plat (RTCA-DP) xCELLigence System (ACEA Biosciences) as per the manufacturer guidelines. In brief, 100 μl of serum-free medium containing 10,000 cells were loaded into the upper wells of CIM-Plate (AGILENT, 5,665,817,001), while 200 μl of complete media were added to the lower chamber. 20 μl of Matrigel (BD Biosciences) basement membrane matrix was used to coat the microporous membrane for invasion or without for migration. For the proliferation assay, E-plate (AGILENT, 5,469,830,001) was used and 100 μl of complete medium containing 10,000 cells was loaded into each well. The plates were incubated for 30 min and then was inserted into the RTCA-DP instrument inside a humidified, 37 °C, 5% CO2 incubator. The obtained data were analyzed by the RTCA software.

Real-Time Cellular Functional Assays

Cell invasion, migration and proliferation capacities were assessed using the Real-Time Cell Analyzer–Dual Plat (RTCA-DP) xCELLigence System (ACEA Biosciences, Santa Clara, CA, USA) as per the manufacturer’s recommendations. In brief, cells (105) in 100 μL of serum-free medium were loaded into the upper wells of CIM-Plate, while complete media (200 μL) were added to the lower chamber. For invasion, the microporous membranes were coated with matrigel (BD Biosciences, Franklin Lakes, NJ, USA) (20 μL). E-plate was used for the proliferation assay. The plates were incubated for 30 min at 37 °C in a 5% CO2 incubator and then were inserted into the RTCA-DP instrument inside the same incubator. The RTCA software (1.2.1) was utilized for data analysis.
