Quantifying miR-181-5p Levels in ADSCs, Exosomes, and BMDMs

The levels of miR-181-5p in ADSCs, exosomes derived from ADSC, and exosome-treated BMDMs were detected by real-time PCR experiment. In brief, total RNA was isolated from exosomes by using the TRIzol solution, followed by reverse transcription to cDNAs by an Easy Script kit (TransGen, China). The relative expression of miR-181-5p was measured by using a SYBR Green SuperMix kit (TransGen) in ABI Prism 7900 (Applied Biosystems, USA) and normalized to the internal control U6. Quantifying miR-301a-3p and STAT3 in ASMCs, ADSCs, and Exosomes

To quantify the levels of miR-301a-3p and STAT3 in ASMCS, ADSCs and exosomes, total RNAs were extracted by using Trizol reagent (Beyotime), reverse-transcribed to cDNA by using Easy Script kit (TransGen, China), and subjected to real-time PCR with a SYBR Green SuperMix kit (TransGen). The quantification of miR-301a-3p and STAT3 was normalized to internal control β‐actin and U6, respectively. Primers were listed in Table S2.
