PCNA Immunohistochemistry Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Tumors were fixed with 10% neutral buffered formalin for 48 h, paraffin-embedded, and cut in 5-μm sections. Heat-induced antigen retrieval was performed in citrate buffer, and after blocking endogenous peroxidase, slides were incubated with primary antibody anti-PCNA antibody (PC10) (1:200 dilution, sc-56, Santa Cruz Biotechnology) as previously indicated [26 (link)]. Then, slides were incubated with a biotinylated antibody and developed with the ABC-HRP Kit (Vector Laboratories, Burlingame, CA, USA) and peroxidase substrate DAB (Sigma-Aldrich, St. Louis, MO, USA). Afterward, slices were examined with a Zeiss Axioplan microscope equipped with a Nikon DXM1200F digital camera. The PCNA cell count was quantified in four randomly selected fields from each animal and analyzed using ImageJ 1.54e software.

Immunohistochemical Analysis of Liver Tissue

Livers were fixed and paraffin embedded. Sections were routinely stained with Hematoxylin&Eosin (7-μm) or incubated with mAb anti-PCNA antibody (PC10) (1:200 dilution, sc-56, Santa Cruz Biotechnology) as previously indicated [53 (link)]. The slices were examined with a Zeiss Axioplan microscope equipped with a Nikon DXM1200F digital camera. The PCNA cell count was quantified in four randomly selected fields from each animal and analyzed using ImageJ software. Ki-67 staining was performed using a specific antibody (sc-23900, 1:200 mouse) from Santa Cruz Biotechnology.

Liver Histology and Immunohistochemistry

Check if the same lab product or an alternative is used in the 5 most similar protocols
Livers were fixed and 7-μm sections were routinely stained with H&E or paraffin sections (5-μm) were prepared following standard procedures22 (link)23 (link)26 (link). The antibodies used were rat mAb anti-CD34 (1:50 dilution, ab8158, Abcam), goat polyclonal anti-antiogenin (C1) (1:200, sc-74528, Santa Cruz Biotechnology), mAb anti-PCNA antibody (PC10) (1:200 dilution, sc-56, Santa Cruz Biotechnology) and anti-CD34 (1:100 dilution, sc-18917, Santa Cruz Biotechnology). The slices were examined with a Zeiss Axioplan microscope equipped with a Nikon DXM1200F digital camera. PCNA index was quantified in four randomly selected fields from each animal, and CD34 positive areas analyzed using ImageJ software.

Apoptotic Cell Quantification in Liver Tissues

Check if the same lab product or an alternative is used in the 5 most similar protocols
Liver and tumor samples were fixed and 5-μm sections were prepared following standard procedures. The slices were examined with a Zeiss Axioplan microscope equipped with a Nikon DXM1200F digital camera. Apoptotic cells with fragmented nuclei were detected in paraffin samples using TUNEL labeling containing fluorescein-dUTP and -dNTPs (TUNEL Label Mix, Roche). TUNEL positive cells were observed and quantified using a NIKON Eclipse E-100 microscope.

Observing halophilic cell growth

Check if the same lab product or an alternative is used in the 5 most similar protocols
WCL brine samples were kept in the laboratory at room temperature and exposed to natural light. Cells were observed to slowly grow under these conditions. Subsamples of these brines were inoculated in a classical culture medium for halophiles (Rodriguez-Valera et al., 1980) slightly modi ed to match the pH and Mg concentrations of the WCLs (Tables S2-S3). Growth was monitored by optical microscopy using a Zeiss Axioplan microscope equipped with a Nikon Coolpix B500 color camera for image acquisition.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
