Trp53loxp

Manufactured by Jackson ImmunoResearch

The Trp53loxP/+ is a laboratory mouse strain that carries a loxP-flanked (floxed) Trp53 allele. Trp53 is the gene that encodes the p53 protein, a key regulator of cellular processes. This mouse model allows for the conditional deletion or modification of the Trp53 gene in a cell-type or tissue-specific manner using Cre recombinase technology.

All procedures and experiments were performed according to the guidelines of the Canadian Council on Animal Care and approved by the animal Care Committee of the Montreal Children's Hospital. WT CD1, mT/mG Gt(ROSA)26Sortm4(ACTB-tdTomato,-EGFP)Luo/J, (Soriano, 1999 (link)), Wnt1-Cre2 (Lewis et al., 2013 (link)) and β-actin-Cre (Lewandoski et al., 1997 (link)) mice on the C57BL/6J genetic background were purchased from The Jackson Laboratory. The R26R strain [Gt (ROSA)26Sortm1Sor (Muzumdar et al., 2007 (link))] on the mix C57BL/6J;129/S4 genetic background was a kind gift from Dr Nagano (Department of Obstetrics and Gynecology, McGill University, Montreal, Canada). All strains were maintained on the CD1 genetic background. The Trp53tm1brn mouse line with loxP sites flanking exons 2-10 of the Trp53 gene was purchased from The Jackson Laboratory (Trp53loxP/+) (stock #008462) (Marino et al., 2000 (link)).

Mice were maintained in the specific pathogen-free (SPF) facility of KAIST Laboratory Animal Resource Center and housed under a 12h/12h light/dark cycle, with free access to regular chow and water. All mice were grouped by no more than 5 mice per cage. Mouse experiments were performed with the approval of the IACUC of Korea Advanced Institute of Science and Technology (KAIST) (KA2020-115). LSL-K-rasG12D, Trp53LoxP, and R26-tdTomato mice were purchased from The Jackson Laboratory and described previously.49 (link),50 (link),51 (link)
Prmt1fl/fl mice (Mouse Genome Informatics [MGI]:4432476) were purchased from the International Mouse Phenotyping Consortium (IMPC) and described previously.13 (link)
Pdx1-CreERT2 mice were provided by Dr. Douglas A. Melton (Harvard University) and described previously.52 (link) Inducible Cre-mediated gene deletion was achieved by i.p. injection of tamoxifen (Sigma) at a dose of 200 mg/kg bodyweight on three alternate days at 3 to 4 weeks of age. tamoxifen was dissolved in corn oil (Sigma) at a concentration of 20 mg/mL.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
