Methamphetamine-Induced Rotational Behavior

Check if the same lab product or an alternative is used in the 5 most similar protocols
At 12–14 days following surgery, rats received an intraperitoneal injection of (+)-methamphetamine hydrochloride (M8750 Sigma) (3 mg/kg), and were then placed in a clean open field container (45 × 45 cm) surrounded by plexiglass walls in a quiet, well-lit room. At 30 min after injection, their activity was recorded for a 10-min period. For scoring, three investigators blind to treatment regimen counted the number of 360-degree ipsiversive rotations, as previously described.22 (link) The container was sanitized with 70% ethanol between each trial and allowed to air dry.

Methamphetamine-induced Behavioral Sensitization in Rats

Check if the same lab product or an alternative is used in the 5 most similar protocols
Male Wistar rats (220±10 g) were housed in a 12 hr light/dark cycle at 22 to 24°C, with food and water ad libitum. All of the experimental procedures were conducted in accordance with the National Institutes of Health’s Guide for the Care and Use of Laboratory Animals. All efforts were made to minimize the number of animals used and their suffering. Methamphetamine hydrochloride (Sigma–Aldrich, M 8750) was dissolved in 0.9% saline. The rats were chronically treated with subcutaneous injections of METH (2 mg/kg), once a day for 5 days, as described previously (15 (link)). Rats become sensitized to METH during 5 days. Normal saline solution was similarly injected into control rats. This dose of METH shows no neurotoxicity but produces behavioral sensitization after repeated treatment in rats (16 (link)).

Apomorphine-Induced Rotational Behavior

Apomorphine-induced circling test was used to reflect DA denervation in rodents with unilateral nigrostriatal lesion50 (link),52 (link). In brief, 14 days after surgery, rats were injected intraperitoneally with apomorphine (M8750 Sigma, 5 mg/kg) and placed in a clean open arena (120 × 120 cm). The whole experiment was performed in a quiet, well-lit room. Thirty minutes after the injection, the rats were recorded for 10 min. The number of rotations towards the contralateral side of lesion was counted.

Pharmacological Interventions in Preclinical Neuroscience

Check if the same lab product or an alternative is used in the 5 most similar protocols
All drugs were dissolved or diluted in 0.9% SAL and administered at a volume of 0.01 ml/g. MDMA (7.5 mg/kg; Organix Inc., O-8571) and METH (2 mg/kg; Sigma-Aldrich M8750) were administered intraperitoneally. The dose of METH was selected on the basis of previous studies (18 (link)), which found that this dose (2 mg/kg) was sufficient to induce locomotor sensitization and conditioned place preference, but not an effect in the three-chamber social preference assay. MOR (10 mg/kg; Sigma-Aldrich, M8777, MOR sulfate salt pentahydrate) and meloxicam (10 mg/kg; VetOne, Ostilox, 5 mg/ml solution) were administered subcutaneously. For TRAP experiments, 4-OHT (Sigma-Aldrich, H6278) was prepared in a mixture of castor and sunflower oil (20:80 ratio) and injected intraperitoneally at a dose of 50 mg/kg.

(+)-Methamphetamine hydrochloride (Sigma-Aldrich, M8750) was freshly dissolved in saline before use and administered subcutaneously (9 mg/kg, s.c.) at the dose 9 mg/kg. The rats were allowed to survive 3 h after the METH administration [21 (link)–23 (link)].

Methamphetamine and Norepinephrine Effects

Check if the same lab product or an alternative is used in the 5 most similar protocols
Reagent doses were optimized in dose-response experiments in both macrophages and BAs, to account for viability and biological effects. (+) Methamphetamine hydrochloride (M8750 – Sigma-Aldrich) was added to the culture media at 60 μM, as previously reported [6 ,7 ,23 ] and cells were harvested at indicated time-points. (−) Norepinephrine (A7257, Sigma Aldrich) was added to the cultures at 0.1 μM. N-acetyl cysteine was added at 5 pg/ml (A7250, Sigma Aldrich). ROS measurements were performed at 30 min and 24 h after stimuli. Assays were performed 24 h after stimulations unless stated in figure legends. Trypan blue (Sigma Aldrich) dye exclusion staining was performed to count cells and assess viability upon plating and retrieval. Cultures with less than 80% viability were discarded.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
