Colorimetric Quantification of Testicular Iron

The content of Fe³⁺ and Fe²⁺ in testicular tissues was detected using an iron colorimetric assay kit (E-BC-K139-S, Elabscience, Wuhan, China) and ferrous iron colorimetric assay kit (E-BC-K773-M, Elabscience, Wuhan, China) according to the manufacturer’s instructions. Testicular tissues were taken and rapidly placed in ice-cold saline to prepare tissue homogenates, then centrifuged at 12,000× g for 10 min at 4 °C. The supernatant obtained was used to measure various indicators. The relative content level of Fe³⁺ and Fe²⁺ was calculated and normalized to mean control levels. Analyses were performed in triplicate (n ≥ 3); the cv values are shown in Supplementary Table S1.

Quantification of Cartilage Biomarkers in OA

After morphological and histological division of intact and damaged areas, human OA cartilage were cut into small pieces and ground into powder under liquid nitrogen condition. Then, we homogenate the cartilage powder with cold normal saline and extracted the supernatant after centrifugation (12,000 rpm, 20 min, 4 °C). Hippocampal Iron and Oxidative Stress
