Renal Function Assessment Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Renal function parameters were assessed by determination of urinary creatinine, serum creatinine, and albumin excretion by commercially available ELISA kits. For quantification of albumin excretion, the Mouse Albumin ELISA Kit (Assay Pro, St. Charles, MO, USA) was used according to the manufacturer’s instructions. Creatinine levels in serum and urine were determined with the Creatinine Urinary/Serum Assay Kit (Cayman Chemical Co., Ann Arbor, MI, USA) according to the protocol provided by the manufacturer. Urea in the serum was determined with the QuantiChromTM Urea Assay Kit (BioAssay Systems, Hayward, CA, USA) according to the protocol provided by the manufacturer.

Urinary Albumin Analysis in Mice

Check if the same lab product or an alternative is used in the 5 most similar protocols
Mice were individually housed in metabolic cages during the last 3 days of treatment, and after 3 h of habituation, 24-hour urine samples were collected. Animals continued to have free access to water and standard laboratory diet during this period. The 24-hour urine samples were stored at −70°C until analysis. Urinary albumin concentration was measured using a mouse albumin ELISA kit (Assaypro, USA) following the manufacturer's protocol.

Urinary Biomarker Measurement Protocol

Check if the same lab product or an alternative is used in the 5 most similar protocols
Urine albumin was determined in the final 3 days of experimentation according to our established procedure [16] (link). Briefly, urinary albumin concentration was measured using a mouse albumin ELISA kit (Assaypro, USA) following the manufacturer’s protocol. Urinary creatinine was determined with an Automatic Analyzer Model 7170 (Hitachi Co, Ltd., Japan). Serum creatinine, blood urea nitrogen, blood uric acid, and albumin/creatinine ratios were all assayed by the Laboratory Department of Xiangya Hospital, Central South University.

Quantifying Secreted Proteins in Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
To assess AFP and albumin secretion, the Mouse alpha Fetoprotein ELISA Kit (Abcam) and the Mouse Albumin ELISA kit (AssayPro), respectively, were used according to the manufacturers' instructions (Table S2, part 2). Supernatants were collected 24 h following the most recent medium change.

Renal Function and Albumin Excretion Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
Renal function was assessed by determination of creatinine clearance. Therefore creatinine in serum and urine was measured according to standard procedures using a Cobas analyzer (Roche, Mannheim, Germany) in the Institute of Clinical Biochemistry and Pathobiochemistry of the University of Würzburg. For quantification of albumin excretion, the Mouse Albumin ELISA Kit (EMA 3201-1, Assay Pro, St. Charles, USA) was used according to the manufacturer's protocol. The absorbance was read at 450 nm with an ELISA reader (Spectra MAX 340, Molecular Devices, Ismaning, Germany) and a standard curve was generated on each plate.

Renal Biomarkers for Kidney Injury

Check if the same lab product or an alternative is used in the 5 most similar protocols
Renal function was assessed by measuring serum creatinine, calculating creatinine clearance and quantifying excretion of albumin, kidney injury molecule-1 (KIM-1) and neutrophil gelatinase-associated lipocalin (NGAL). Creatinine in urine and serum was determined using creatinine urinary/serum colorimetric assay kits (No. 500701/700460, Cayman Chemical Company, Ann Arbor, MI, USA) according to the manufacturer’s protocol. The mouse albumin ELISA kit (EMA3201-1, Assay Pro, St. Charles, IL, USA), the mouse KIM1/TIM1 ELISA kit (PicoKineTM, EK0880, BosterBio, Pleasanton, CA, USA) and the mouse NGAL ELISA kit (KIT 042, BioPorto, Gentofte, Denmark) were used for the quantification of albumin, KIM-1 and NGAL excretion following the manufacturer’s protocol. Albumin, KIM-1 and NGAL were related to urinary creatinine.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
