Fluorometric intracellular ros assay kit

Manufactured by Merck Group
Sourced in United States

The Fluorometric Intracellular ROS Assay kit is a laboratory instrument designed to measure the levels of reactive oxygen species (ROS) within cells. It utilizes a fluorescent probe that detects the presence of ROS, providing a quantitative assessment of intracellular oxidative stress.

Fluorometric ROS Assay in A549 Cells

Check if the same lab product or an alternative is used in the 5 most similar protocols
The Fluorometric Intracellular ROS Assay kit (Sigma-Aldrich; Merck KGaA) was used to detect ROS produced by H2O2-treated A549 cells, according to the manufacturer's protocol. The assay was performed in 96-well plate and read using a fluorescence microplate reader resulting in a fluorometric product (l ex = 490/l em = 520 nm) proportional to the amount of ROS present.

Intracellular ROS Detection in DEFs

Check if the same lab product or an alternative is used in the 5 most similar protocols
Detection of intracellular ROS in DEFs was performed with a fluorometric intracellular ROS assay kit (Sigma–Aldrich, USA) based on a superoxide fluorescent probe (dihydroethidium). DEFs were plated in 96-well black microplates with clear flat bottoms to a cellular concentration of 1 × 104 cells/mL. The cells were then allowed to adhere for 24 h and infected with DTMUV, and the cells were harvested according to the instructions at 6, 12, 24, 36, 48 and 60 h of infection. The ROS reagent was reconstituted with dimethyl sulfoxide and then diluted in the assay buffer. Shortly afterwards, the ROS reagent solution was added to each well, and the plates were incubated for 1 h. Subsequently, the data were collected using a fluorescence microscope, and the fluorescence intensity was measured in a multifunctional microplate reader (Thermo Scientific, Massachusetts, USA) with an excitation wavelength of 520 nm and an emission wavelength of 605 nm. The data are shown as relative fluorescence compared to the fluorescence of the negative control.

Fluorometric Intracellular ROS Assay

Check if the same lab product or an alternative is used in the 5 most similar protocols
ROS (superoxide and hydroxyl radicals) were detected with the Fluorometric Intracellular ROS Assay Kit (Sigma-Aldrich, USA) following the manufacturer’s protocol using fluorometric sensors localized to the cytoplasm, resulting in a fluorometric product proportional to the amount of ROS present. The fluorescence intensity was determined at λex = 540 nm and λem = 570 nm. Fold increases in ROS levels were determined by comparison with the control group.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
