Subcutaneous Tumor Growth Assay in M-NSG Mice

Four to six weeks old female M-NSG mice (Shanghai Model Organisms Center, Shanghai, China) were used for experiments. The mice were randomly divided into 4 groups. Bel-7402 cells after lentivirus infection were subcutaneous injected into the right side of the mice with the density of 5 × 106 and the weights, tumor numbers as well as the tumor size recorded until the mice were sacrificed. Tumor size (mm3) was measured with vernier calipers and calculated with Eq. (1): Tumorsize=(W2×L)/2
in which W is the width and L is the length. The animal experiment was approved by the Institutional Animal Care and Use Committee in Zhejiang University, and was carried out following the institutional guidelines. The IACUC number of the animal experiment was IACUC-s21-027.

Xenograft Model for Tumor Immunotherapy

Female NOD-PrkdcscidIl2rgem1Smoc (M-NSG) mice aged 6 weeks old were purchased from Shanghai Model Organisms Center (Shanghai, China). NSG mice were intraperitoneally injected with 1×107 freshly isolated PBMCs from healthy donors, and 2×106 SGC-7901 cells were implanted subcutaneously into the right flank of the mice. After 8 days, the mice were treated with PBS (mock), RCAd, RCAd-shPD-L1, RCAd-LTH, or RCAd -LTH-shPD-L1. Tumor volumes and body weights were recorded every 2 days.
