Before treatment with PNF and piperine, the 3T3 L1 cells were starved with serum-free DMEM (high glucose) media for 16–18 h. After 30 min of pretreatment with PNF and piperine, the cells were treated with eugenol for 7 min and then lysed with 0.1 M HCl. cAMP levels were measured using the Direct cAMP EIA Kit (Enzo Life Sciences).

Bordetella pertussis ACT-induced cAMP Assay

cAMP produced by ACT-coated bacteria (Bordetella pertussis 18323 H-) (incubation of 5–100 μg ACT/106 bacteria) was assayed for 10 min at 37 °C with 2 nM CaM in AC reaction buffer (30 mM Tris-HCl, pH 7.4, 20 mM MgCl2 and 100 μM CaCl2), then the reaction was started by addition of 5 mM ATP. After 10 min at 37 °C with continuous stirring the reaction was stopped with 0.1 M HCl. The cAMP production was calculated by the direct cAMP EIA kit (Enzo lifesciences).
