Silencing Porcine STING with siRNA

All siRNAs targeting porcine STING (mRNA accession number: NM_001142838.1) were designed and synthesized by Invitrogen (ThermoFisher Scientific) and the siRNA sequences are listed in Table 3. CD163-PAMs were plated into 24-well plates and the cells with 70–80% confluency were transfected with 30–50 nM of siRNA using Lipofectamine 2000 according to the manufacturer’s instructions. To determine the efficiency of the knockdown, total RNA was extracted from the cells at 24 h post transfection, and endogenous mRNA levels of STING were quantified by RT-qPCR. In parallel, cells were lysed, and proteins were measured by Western blotting for STING protein expression with polyclonal anti-STING rabbit antibody (1:2000) (Proteintech, Rosemont, IL, USA).

Adipose Tissue Protein Analysis

White Adipocytes were isolated from eWAT as the floating fraction and lysed in RIPA buffer (CST, 9806). Protein concentration in whole cell lysis was quantified using BioRad Protein Assay Kit (BioRad, 5000201EDU). 3mg of total proteins were loaded on 4~20% MINI PROTEAN Gels and probed by indicated antibodies.
