Quantitative Western Blot Analysis of NSCs

Cultured NSCs were washed with ice-cold PBS and lysed with 2 × SDS lysis buffer. Protein concentrations were determined by the BCA protein assay kit (Pierce, Rockford, IL, USA). Proteins were separated on 8% SDS-polyacrylamide gels (Bio-Rad Laboratories, Inc., Hercules, CA, USA) and transferred to a PVDF membrane (Millipore, Billerica, MA, USA). Membranes were blocked in 5% non-fat milk powder in TBS-T (0.1% Tween-20 in TBS), and incubated with primary antibodies overnight at 4°C. After washing in TBS-T, membranes were incubated with HRP-conjugated secondary antibodies (Invitrogen) for 1 h at room temperature with signals detected using ECL Super Signal (Pierce). Quantifications were done using Image-Quant Software (Molecular Dynamics, Sunnyvale, CA, USA). The primary antibodies used include rabbit anti-p16 (Abcam, Cambridge, MA, USA), anti-p21 (Abcam), anti-ku70 (Abcam), anti-Xrcc2 (Abclonal Technology, Wuhan, China), anti-Xrcc3 (Abclonal Technology), anti-Xrcc4 (Abclonal Technology) and mouse anti-βactin (Sigma).
