Rejuvenating Old Mice Skin

Male C57BL/6 mice were purchased from the Sankyo Labo Service Corporation, Inc. (Tokyo, Japan). Young (15 weeks old) and old (90 weeks old) mice were both used in this study. The complexes were prepared by combining invivofectamine 2.0 (Life Technologies, Carlsbad, CA, USA) and SFRP4 siRNA (152089, 152090, and 152091; Silencer™ siRNA, Thermo Fisher Scientific), according to the manufacturer’s instructions. The SFRP4 siRNA complex, negative siRNA (Silencer™ Negative Control No. 1 siRNA, Thermo Fisher Scientific) complex, or control (PBS) were injected at 4 mg/kg into old mice by intravenous injection weekly (1.5 mg/kg in week 1 and 1 mg/kg in weeks 2 and 3). Experiments were performed with n = 5 for each group. One week later, the skin was collected in whole layers, and tissue specimens were fixed by immersion in 4% paraformaldehyde, embedded in paraffin, sectioned, and stained with hematoxylin-eosin and Masson’s trichrome staining. Tissues for RNA recovery were later immersed in RNA (Qiagen, Hilden, Germany) and stored at −20° C until use.

Male C57BL/6 Mice Housing Protocol

Male C57BL/6 mice (8 ± 1 weeks old) were purchased from Sankyo Labo Service Corporation, Inc. (Tokyo, Japan). Animals were reared under normal environmental conditions; lighting was maintained on a 12 h light-dark cycle, with ambient temperature maintained at 25 ± 2°C. All animal experiments were approved by the Institutional Animal Care and Use Committee of Josai University (approval number: H24011-2012/04/04).

Chronic Liver Injury Mouse Model

Male C57BL6 mice (8-week-old) weighting 24–26 g were purchased from Sankyo Labo Service Corporation, Inc. (Tokyo, Japan). Mice were maintained for a week before starting experiment in environment with constant light/dark cycle (12 h), ambient temperature (23 ± 2 °C), and humidity (40%). During experiments, they were maintained in the same condition. ISO (Kanto Chemical Co., Inc., Tokyo, Japan) was dissolved in phosphate-buffered saline (PBS) at a concentration of 5 mg/ml and subcutaneously administered to mice (25 mg/kg) daily for a week41 (link). A DDC-containing diet was made by mixing 0.1% DDC (Sigma-Aldrich) in MF diet (Oriental Yeast Co. Ltd., Tokyo Japan). To induce chronic liver injury, mice were fed DDC-diet ad libitum for 14 days. During DDC-feeding, ISO was further administered every 5 days. Mice were euthanized by isoflurane for sample collection. All animal experiments were performed under approval from the Animal Experiment Committee of Sapporo Medical University. The animal studies are reported with the recommendations in the ARRIVE guidelines. All the relevant institutional guidelines, along with the ARRIVE guidelines were followed during the study.

Modulating Gut Microbiome in Mice

Male C57BL/6 mice were purchased from the Sankyo Labo Service Corporation (Tokyo, Japan) and were kept under conventional conditions. Male MCH (ICR) mice, germ-free (GF) male ICR mice, and ICR-derived inbred strain IQI mice were purchased from CLEA Japan, Inc. (Tokyo, Japan). GF male MCH and IQI mice were maintained in vinyl isolators. Male MCH mice were kept under specific pathogen-free (SPF) conditions. These mice were fed γ-ray-sterilized AIN-93G (Oriental Yeast, Tokyo, Japan). In order to evaluate the effect of sugar alcohol on diarrhea, mice were administered water drinking water containing 5% or 10% sorbitol, or 5% mannitol for 4 days. For experiments involving antibiotics, mice were administered drinking water containing ampicillin (1 g/L), streptomycin (5 g/L), erythromycin (200 mg/L), or vancomycin (250 mg/L). For gnotobiotic experiments, GF male ICR or IQI mice (obtained from CLEA Japan) were bred in vinyl isolators and fed γ-ray-sterilized CMF. During the experiment, GF mice were fed a γ-ray-sterilized AIN-93G.
All mice were housed under 21–22 °C with a 12 h alternating light–dark cycle at the animal facilities of Faculty of Pharmacy, Keio University (Tokyo, Japan). All animal experiments were performed according to the Institutional Guidelines for the Care and Use of Laboratory Animals in Research and were approved by the local ethics committees at Keio University.
