Sections (4 µm thick) were deparaffinized. Immunohistochemical staining was performed using a Ventana Discovery XT instrument supplied by Ventana Medical Systems Inc. (Tucson, AZ, USA) according to the manufacturer’s instructions. The primary antibodies against TTF-1 (SP141) were purchased from Ventana Medical Systems Inc. (Catalog No. 790-4756, ready to use without dilution), and thyroglobulin was purchased from Dako (Catalog No. M078101, Glostrup, Denmark, ready to use without dilution). The reactions were visualized with 3,3′-diaminobenzidine followed by counterstaining with hematoxylin.

Thyroid Signaling Pathway Analysis

Five-month-old animals were sacrificed and TdTomato-positive thyroid tissue was dissected under a stereomicroscope and homogenized in 40 µl RIPA buffer (Pierce, Waltham, MA). Thyroid Carcinoma Cases with CRCC-like Appearance

We reviewed 12,064 primary thyroid carcinoma cases resected in Kuma Hospital from 2005 to 2015, and selected two cases (cases 1 and 2) with CRCC-like appearance. CRCC-like appearance was defined as a prominent cell border, perinuclear clear cytoplasm, a wrinkled nuclear membrane, and binucleation. One case (case 3) from Fukushima Medical University School of Medicine with similar histological findings was also included in this study.
