Solubility Characterization of Test Compound

Test compound (2 mg) was suspended with 1 mL of normal saline, pure water, or 5% glucose aqueous solution in glass tube, and the suspension was shaken at 150 rpm for 48 h at 25 °C. An aliquot was filtered by PVDF filter unit (Mini-Uni PrepTM, GE Healthcare), and the filtrate was diluted with pure water. The diluted sample solution was injected into HPLC system (column (Mightysil RP-18, Kanto Chemical Co. Inc.), UV/vis detector (UV-2077, JASCO), pump (PU-2089, JASCO), and column oven (CO-965, JASCO)) and the peak area was recorded at 254 nm. The concentration of solution was calculated with a calibration curve. Examinations of all conditions were performed three or four times.

Rubisco Quantification by SEC-HPLC

Ultracentrifuged juices were filtered through Mini-UniPrep TM filters with pore size 0.2 μm (Whatman TM , GE Healthcare Life Sciences) before injection into a HPLC (Agilent, United States) with a Yarra 1.8 µm SEC-X150 column (size: 150 x 4.6 mm) (Phenomenex, United States). Four μL of juices were injected into the HPLC and compared to a D-Ribulose 1,5-diphosphate carboxylase (Rubisco) standard from spinach (Sigma-Aldrich, Germany). Protein was eluted with a 0.05 M PO4 buffer with 0.3 M NaCl and pH 6.8 at a flow of 0.3 mL min -1 . UV absorbance was measured at 214 nm. Size exclusion analysis was performed in triplicate.
