Plasma Resistin Quantification

Plasma resistin levels were measured by an enzyme-linked immunosorbent assay (ELISA) kit (Abcam ab205574, detection limits 31.25-2000 pgÁml -1 ). For this purpose, 0.5 ml of fasting venous blood was collected in K2-coated tubes containing 30 μl (0.5 molÁL -1 ) of EDTA plus 2000 KIU of aprotinin. Samples were mixed and centrifuged at 2000 g for 20 min. After centrifugation, 2 ml of cold acetone (4 C) was added to 0.5 ml of plasma, mixed and centrifuged at 2000 g for 20 min at 4 C. The supernatant was added to 4 ml of cold petroleum ether, mixed, centrifuged and dried under a vacuum to remove any residual acetone. Samples were then stored at À20 C until use. Aliquots of plasma were diluted 1:50 and used for the assay.
Plasma resistin levels were expressed as nanograms per millilitre.
