HPLC Analysis of Isoflavonoids

Samples were analyzed using a reversed-phase HPLC system (Shimadzu, Japan) with a Zorbax Eclipse XDB-C18 column (5 μm × 4.6 mm × 250 mm) (Agilent Technologies, USA). Chromatography was performed at room temperature (RT) at a flow rate of 0.5 mL/min, and 10 μL was analyzed for 110 min. The mobile phase consisted of 0.1% formic acid (A) and acetonitrile (B) in a ratio specified by the following binary gradient with linear interpolation: 0 min 20% B, 60 min 30% B, 70 min 60% B, and 100 min 70% B. The column eluent was monitored by UV spectrophotometry for absorbance at a wavelength of 280 nm. The contents of genistein, prunetin, and sakuranetin (Sigma) in chloroform fraction were quantified.

Phytochemical Profiling of Rhus Species

Sakuranetin (Biomarker; purity: ≥95%), rutin (antioxidant standard; purity: ≥94%), Tween_40, 2,2-diphenyl-1-picrylhydrazyl (DPPH), β-carotene, linoleic acid were purchased from Sigma Aldrich (USA). The developed HPTLC plates were then scanned and documented by CATS 4 (CAMAG) and TLC Reprostar 3 (CAMAG), respectively.

Astragalin, apigenin, naringenin, kaempferol-3-rutinoside, and tiliroside were supplied by Roth (Karlsruhe, Germany). LC–MS grade water was prepared using a Millipore Direct-Q3 purification system (Bedford, MA, USA).

Synthesis of Sakuranetin from Naringenin

Sakuranetin Cytotoxicity Assay

Cell viability was determined using the MTS (3-(4,5-dimethylthiazol-2-yl)-5-3(carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium) reduction method (CellTiter 96 One Solution Cell Proliferation Assay Kit, Promega, Madison, WI, USA), based on the measurement of mitochondrial respiration in living cells. Optical density was measured at 490 nm with a microplate reader (Molecular Devices, Sunnyvale, CA, USA).
