The E171 food additive was purchased as powder from the website of a French commercial supplier of food colouring agents. These food-grade TiO2 particles were prepared following the generic Nanogenotox dispersion protocol as previously described [7 (link)]. The E171 stock suspension (25.6 mg/mL) was sonicated in an ice bath for 16 min at 30% amplitude (VCX 750-230 V, Sonics Materials) to obtain a stable dispersion of TiO2 particles and then stocked at 4 °C during 15 days maximum before use. Dynamic light scattering (DLS, ZetaSizer nano ZS; Malvern Instruments Ltd.) measurements were performed on TiO2 particles in ultrapure water (pH = 8.92) and perfusion medium (PM = Earle + 2% BSA, pH 7.4). Five μL of E171 stock suspension were diluted in 3 mL of PM and the hydrodynamic diameter (Z-average), polydispersity index and zeta potential of the TiO2 particles were measured. Particle diameters were measured by scanning electron microscopy (SEM,n = 600 particles) and are expressed as the percentage of NPs by number. The specific surface area (SSA) of the particles was assessed according to the Brunauer, Emmet and Teller method (BET).

Characterization of Food-Grade Silica Nanoparticles

The SiO2 sample was purchased as powder from a French commercial supplier producing food-grade ( fg-SiO2 ) precipitated amorphous silica (INS 551) in the frame of the PAIPITO project funded by the French National Research Agency.17 (link) For physicochemical characterization, fg-SiO2 was sonicated (VCX 750-230V; Sonics and Materials, Inc.) in ultrapure water ( 1mg/mL ) for 1 min at 40% amplitude in an ice bath.24 (link) The hydrodynamic diameter (Z-average), polydispersity index, and zeta potential were determined using dynamic light scattering (DLS) (Zetasizer Nano ZS; Malvern Instruments Ltd.) with 10μL of fg-SiO2 suspension diluted in 2mL of ultrapure water (Table S1).24 (link) Transmission electron microscopy (TEM) analysis was performed on a JEM 1400 (JEOL) equipped with camera Orius SC1000B (Gatan, Inc.) driven by DigitalMicrograph software (Gatan-Ametek). The sonicated suspension of fg-SiO2 was diluted in ultrapure water ( 10mg/mL ), and size measurement was determined from bright-field TEM images by using the image processing open-source software ImageJ (NIH).
