LUAD Pathology: Correlating mRNA Levels

To assess correlations among mRNA levels, we collected pathologically confirmed LUAD samples from 59 patients who underwent resections at the Johns Hopkins Hospital (Baltimore, MD, USA), the Johns Hopkins Bayview Medical Center (Baltimore, MD, USA), and the Medical College of Wisconsin Froedtert Memorial Hospital (Milwaukee, WI, USA) (20 (link)). Formalin-fixed paraffin-embedded TMA sections were constructed from blocks of resected specimens from 165 LUAD patients who were treated at the Sacro Cuore Don Calabria Hospital (Negrar, Verona, Italy) (21 ). None of the patients received any therapies before their resections. Antibodies for YAP1 (EP1674Y; Abcam, Cambridge, UK; #ab52771; 1:300) and pSTAT3 (D3A7; Cell Signaling Technology; #9145; 1:25) were used for IHC, which were scored blindly by one pathologist (Dr Gabrielson). Intensity scores (0; none, 1+; weak, 2+; moderate, 3+; strong) of nuclei staining were evaluated by light microscopy; a score of 0 was considered negative, and scores of 1+, 2+ or 3+ were considered positive.

Western Blot Analysis of Protein Expression

Cells were harvested by scraper and lysed with RIPA buffer supplemented with 1× complete protease inhibitor cocktail (Roche) following the manufacturer’s protocol. Protein concentrations were determined using a Pierce BCA protein assay kit (Thermo Scientific). Lysates were separated on SDS-PAGE gels and transferred. Membranes were immunoblotted with the following antibodies: YAP (ab52771, EP1674Y, abcam; 1:1000), TRAM2 (ab109176, EPR2658, abcam; 1:5000), FSTL-1 (ab11805, abcam, 1:1000), Vimentin (550,513, BD Biosciences, 1:2000), HSP90 (610,418, BD Biosciences, 1:5000), and GAPDH (sc-32,233, Santa Cruz, 1:2000). Protein bands were visualized using corresponding secondary antibodies (Dako) and ECL reagent (GE Healthcare).
