Recombinant human pcsk9

Manufactured by R&D Systems
Sourced in United States

Recombinant human PCSK9 is a protein produced in a laboratory setting. It is a member of the proprotein convertase subtilisin/kexin family and is involved in the regulation of low-density lipoprotein (LDL) cholesterol levels.

Recombinant human PCSK9, mouse monoclonal anti-PCSK9, and polyclonal goat IgG were from R&D Systems/BioTechne, Minneapolis, MN, USA. Anti-human CD62P-PE (clone AK4) and anti-human CD42b-APC (clone HIP-1) were from Biolegend (San Diego, CA, USA). LDL was purchased from Kalen Biomedical (Germantown, MD, USA). Mouse control IgG was procured from Santa Cruz Biotechnologies (La Jolla, CA, USA). Repatha (evolocumab) was from Amgen (Thousand Oaks, CA, USA) and conjugated with fluorescein isothiocyanate (FITC, Sigma-Aldrich/Merck, Darmstadt, Germany) by standard methods.

PCSK9 Regulation of CaCo-2 Polarization

Check if the same lab product or an alternative is used in the 5 most similar protocols
Cultured CaCo-2 cells were obtained from American Type Culture Collection (ATCC, VA). CaCo-2 cells were first grown for 3 days in EMEM specialty medium (ATCC, VA) in 20% FBS supplemented with 1% penicillin-streptomycin, at 37°C, 5% CO2, until they reached 80–90% confluence. Induction of cell polarization is necessary for CaCo-2 cells to resemble human intestinal enterocytes in vivo. To induce polarization, CaCo-2 cells were maintained in culture in 10% FBS-DMEM for 14 days in specialized, collagen-treated transwell plates with 0.4 μM pore size polycarbonate filters (CoStar, DC).
Unless otherwise indicated, all studies were performed at least three times. Optimally polarized confluent CaCo-2 cells in 1% FBS-DMEM were treated for 24 hours with recombinant human PCSK9 (R&D Systems, MN) reconstituted in Millipore H20. PCSK9 was mostly used at the concentration of 10 μg/mL for 24 hours13 (link), although many experiments included different doses (0, 5, 7.5, 10 and 12 μg/mL) and different times (0, 2, 4, 8, 12, and 24 hours).

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
