ChIP-qPCR Assay for RUNX1 and Elk-1

ChIP assay was performed as previously described 18 . Briefly, chromatin from crosslinked cells (20 x 10 6 HL-60 cells and 70 x 10 6 in Jurkat cells per condition) was sonicated, incubated overnight with goat anti-RUNX1 (C-19), rabbit anti-Elk-1 (I-20) (Santa Cruz Biotechnology, Inc.) and goat (RUNX1 IP) and rabbit (Elk-1 IP) anti-IgG antibodies (Sigma-Aldrich) in RIPA buffer, and precipitated with protein G/A-Sepharose. Cross-linkage of the co-precipitated DNA-protein complexes was reversed, and DNA was used as a template for quantitative PCR (qPCR). Primers employed are shown in Supplementary Table 3.
