For the co-culture–based bystander experiment, 10.000 HER2-positive SKBR-3 and 2.000 HER2-negative MDA-MB-468 cells were treated with trastuzumab-LP5 DAR8 or Enhertu at concentrations ranging from 0.05 to 3 μg/mL. After 5 days, cells were stained with LIVE/DEAD Fixable Aqua (Thermo Fisher Scientific) and HER2-PE (BioLegend) to distinguish between target-positive and -negative cells. Immunophenotyping of Breast Cancer Cells
