Anti flg

Manufactured by Abcam
Sourced in United States

Anti-FLG is a primary antibody that specifically recognizes the Filaggrin (FLG) protein. Filaggrin is a structural protein that plays a crucial role in the formation of the skin's protective barrier. This antibody can be used in various immunoassay techniques to detect and quantify Filaggrin expression in biological samples.

Dulbecco’s modified Eagle’s medium (DMEM), fetal bovine serum (FBS), phosphate-buffered saline (PBS), trypsin-ethylenediamine tetra acetic acid (EDTA), and streptomycin/penicillin were purchased from Welgene (Daegu, Korea) and Gibco (Grand Island, NY, USA). Anti-DNP-IgE, bovine serum albumin (BSA), p-nitrophenyl-N-acetyl-β-d-glucosaminide, DNCB, and 1-Iodo (1-iodohexadecane) were purchased from Sigma-Aldrich (St. Louis, MO, USA). DNP-BSA was purchased from Thermo Scientific (Waltham, MA, USA) and TNF-α from R&D Systems (Minneapolis, MN, USA). Anti-SNAP23, anti-syntaxin 4, anti-VAMP8, anti-LOR, and anti-FLG antibodies were from Abcam (Cambridge, MA, USA), and the anti-β-actin antibodies were from Sigma-Aldrich. Anti-rabbit IgG HRP-conjugated and anti-mouse IgG HRP-conjugated antibodies were from Cell Signaling (Danvers, MA, USA).

NHEKs were lysed in Radioimmunoprecipitation assay buffer (Millipore). After centrifugation at 13,000 rpm for 10 minutes at 4 C, the supernatant was loaded onto a 5e20% gradient gel and stained with the following antibodies: anti-KRT10 (Biolegend), anti-DSC1 (Novus), anti-FLG (Abcam), anti-glyceraldehyde-3-phosphate dehydrogenase (Santa Cruz Biotechnology, Santa Cruz, CA), antiphosphorylated and total forms of p44/42 MAPK, c-Jun N-terminal kinase, p38, and p65 of the NF-kB (Cell Signaling Technology, Beverly, MA).

PEVs were labeled with DiI (Molecular Probes, Eugene, OR) as described in the Supplementary Material. NHEKs were treated with DiI-labeled PEVs with or without endocytic inhibitors, stained with an anti-early endosome antigen 1 antibody (Abcam, Cambridge, MA), and analyzed under a confocal microscope (LSM 700; Carl Zeiss, Jena, Germany). For immunohistochemical analysis, replicate sections from reconstituted human skin were stained with the following antibodies: anti-KRT10 (Biolegend, San Diego, CA), anti-DSC1 (Novus, Littleton, CO), anti-FLG (Abcam), and anti-Ki67 (Abcam). Detailed protocols for immunological analyses are described in the Supplementary Material.

About PubCompare

Our mission is to provide scientists with the largest repository of trustworthy protocols and intelligent analytical tools, thereby offering them extensive information to design robust protocols aimed at minimizing the risk of failures.

We believe that the most crucial aspect is to grant scientists access to a wide range of reliable sources and new useful tools that surpass human capabilities.

However, we trust in allowing scientists to determine how to construct their own protocols based on this information, as they are the experts in their field.
