Procurement of Human Cancer Tissues

Human primary liver, colon and lung carcinomas, and normal liver, colon and lung tissue specimens surgically removed and snap-frozen in liquid nitrogen were obtained from Alenabio (Xian, China) along with pathologic information (Supplementary Table 4). Protein extracts were prepared from three normal tissues and five tumour tissues. Frozen human cancer and adjacent non-cancer tissue microarrays were purchased from US Biomax (FLV402 for HCC, FCO405a for colon cancer and FLC320 for lung cancer). Paired cancer and adjacent non-cancer paraffin tissue sections (HLiv-HCC180Sur-02 for HCC, HCol-Ade180Sur-03 for colon cancer and HLug-Squ150Sur-01 for lung cancer) were purchased from Shanghai Outdo Biotech (Shanghai, China). All protocols using human specimens were approved by the Institutional Review Board of Chinese Academy of Medical Sciences and Peking Union Medical College. Informed consent was obtained from all subjects. The study conforms to the principles outlined in the Declaration of Helsinki.

Evaluating GSDMD Expression in Lung Cancer

Two commercial tissue microarrays (HLugA180Su02 and HLugSqu150Sur01; Shanghai Outdo Biotech, Shanghai, China) were used to evaluate GSDMD expression in LUAD and LUSC. Antigen retrieval was performed by microwave heating in citrate buffer (pH 6.0) for 5 min. Microarrays were incubated with the primary antibody (GSDMD; 1:100; cat. no. sc-81868; Santa Cruz Biotechnology, Inc., Santa Cruz, CA, USA) at 4°C overnight. Slides were analyzed separately by two independent pathologists. Staining intensity was scored as negative (0), weak (1 (link)), medium (2 (link)) or strong as previously described (3 (link)). An overall GSDMD expression score was calculated by multiplying the intensity and positive percentage scores.

Immunohistochemistry and Immunofluorescence in Lung Cancer

The immunohistochemical staining was performed as described previously19 (link). 55 small cell lung carcinoma tissue samples were from small cell lung carcinoma tissue array LC802 (US Biomax, Rockville, MD) and lung carcinoma tissue array OD-CT-RsLug01-007 (Outdo Biotech, Shanghai, China). 139 paired lung cancer tissues were from human lung cancer array OD-CT-DgLug01-007, HLug-Ade150Sur-01 and HLug-Squ150Sur-01 (Outdo Biotech, Shanghai, China).
