Culturing Glioma Cell Lines

The p53-wild type U87MG glioma cell line was obtained from American Type Culture Collection (LGC Promochem, Teddington, UK). The human p53-mutant T98 and U251 glioma cell lines were obtained from Cell Bank Interlab Cell Line Collection (ICLC, Italy). Glioma cells were grown in Eagle's Minimum Essential Medium (Lonza Bioresearch, Basel, Switzerland) supplemented with 10% (v/v) heat-inactivated fetal bovine serum (FBS), 2mM L-glutamine and 100 IU/ml of penicillin, 100 μg of streptomycin (Lonza), 1 mM sodium pyruvate (Lonza) and non-essential amino acids (Lonza). Cell lines were maintained at 37°C, 5% CO2 and 95% of humidity.

Culturing Glioma Cells and Patient-Derived GSCs

The U87MG glioma cell line was purchased from the American Type Culture Collection (Manassas, VA, USA) and cultured in Dulbecco's Modified Eagle's Medium (DMEM, Lonza, Switzerland) supplemented with fetal bovine serum (10%; Biotechnics Research, Lake Forest, CA, USA), penicillin/streptomycin (1%; Life Technologies, Carlsbad, CA, USA), l-glutamine (2 mmol/L; Life Technologies), and gentamicin (50 µg/mL; Cellgro, Manassas, VA, USA). Patientderived GSCs were kindly provided by Dr Ichiro Nakano. 17 GSCs were cultured in DMEM/F12 (Lonza) supplemented with B27 (0.04%; Invitrogen, Waltham, MA, USA), epidermal growth factor (EGF; 20 ng/mL; R&D Systems, Minneapolis, MN, USA), basic fibroblast growth factor (bFGF; 20 ng/mL; R&D Systems), penicillin/streptomycin (1%), l-glutamine (2 mmol/L), and gentamicin (50 µg/mL).

Ouabain Inhibits U-87MG Glioma Cell Viability

The human U-87MG glioma cell line was obtained from the American Type Culture Collection (Mannasas, VA, USA) and cultured in DMEM containing 10% FBS and 1% penicillin/streptomycin in a humidified incubator of 5% CO2 at 37°C. The cells were randomized into two groupings: Ouabain-treated groups (0.05, 0.25, 0.5, 2.5, 5 and 25 µmol/l for the MTT assay; 0.05, 0.5, 2.5 and 25 µmol/l for other experiments) and a control group (cultured with an equal volume of culture medium containing 0.1% DMSO).
