Membrane Protein Solubilization and Separation

Membrane protein was solubilised in 1× NuPAGE® LDS sample buffer (0.025 mol/L Tris pH 8.5, 0.5% LDS, 2.5% glycerol, 0.05 mol/L dithiothreitol (DTT), 0.05 mol/L SERVA Blue G250, 0.04 mmol/L phenol red). Fifty μg of protein from each sheep was loaded in duplicate lanes on Bolt 4–12% Bis-Tris gels. Proteins were separated in a NuPAGE® MOPS running buffer system (Invitrogen, CA, USA) at a constant 200 V using a mini gel tank (Life technologies, CA, USA). Gels were stained with 0.1% coomassie brilliant blue (CBB R250) in 50% methanol (Chem–supply, SA, Australia) and 10% glacial acetic acid (Chem–supply, SA, Australia), then destained in 40% methanol and 10% glacial acetic acid. Gel images were obtained using a gel doc system (BioRad, CA, USA). Each gel lane was then cut into 9 pieces prior to protein digestion with trypsin and elution from gel piece.

Yoghurt Starter Culture and Probiotics

A commercial yoghurt starter culture (YOFLEX®—consisting of Streptococcus thermophilus and Lactobacillus delbrueckii subsp. bulgaricus) and three probiotic strains (Lacticaseibacillus rhamnosus LGG, Lacticaseibacillus casei 431, Bifidobacterium animalis subsp. lactis BB-12) were kindly supplied by Chr. Pancreatin was purchased from Alfa Aesar (Ward Hill, MA, USA). Mangiferin, pyrogallol, gallic acid, caffeic acid, 3-hydroxy phenyl acetic acid, 3-(2-hydroxyphenyl) propionic acid, porcine pepsin, α-amylase from aspergillus oryzae, p-nitrophenyl-α-d-glucopyranoside, bile salts, LC-MS grade formic acid, acetic acid, 4-methyl valeric acid, guar, casein, tryptone, cysteine HCl, peptone, mucin, pectin, potato starch, yeast extract, and MRS agar were acquired from Sigma–Aldrich (Castle Hill, NSW, Australia). Sodium carbonate (anhydrous), HCl, NaHCO3, NaOH, (NH4)2CO3, KCl, NaCl, KH2PO4, MgCl2(H2O)6, CaCl2, K2HPO4, Na3PO4, MgSO4.7H2O Tween 80, orthophosphoric acid, acetonitrile, and methanol of HPLC grade were acquired from Chem-supply Pty Ltd. (Melbourne, VIC, Australia).

Synthesis and Characterization of Platinum(II) Complexes

All reagents were used as received unless otherwise specified, all organic solvents utilised were of analytical grade, and MilliQTM water (Merck, Melbourne, Australia) was used. Potassium tetrachloroplatinate (K2PtCl4) was acquired from Precious Metals Online. 1S,2S-Diaminocyclohexane (SSDACH, 98%), 1,10-phenanthroline (Phen), 5,6-dimethyl-1,10-phenanthroline (56Me2Phen), silver nitrate, N,N′-dicyclohexylcarbodiimide (DCC), N-(3-Dimethylaminopropyl)-N’-ethylcarbodiimide hydrochloride (EDC-HCl), N-hydroxysuccinimide (NHS), indomethacin and aspirin were purchased from Sigma-Aldrich, Sydney, NSW. Sep-Pak® C18 columns were obtained from Waters, Australia. Methanol, diethyl ether, ethyl acetate, dichloromethane (DCM), dimethyl formamide (DMF), dimethyl sulfoxide (DMSO) and acetonitrile (ACN) were acquired from Chem-Supply, Adelaide, Australia. Hydrogen peroxide (H2O2) (30%) was purchased from VWR chemicals, Philadelphia, United States. Potassium iodide (KI) was obtained from Merck, Melbourne, Australia. Ultra-pure HNO3 (69%) was acquired from Choice Analytical, Sydney, Australia. The certified reference standard that was used for the ICP-MS experiments was purchased from High-Purity Standards, North Charleston, United States. The deuterated solvents deuterium oxide (D2O, 99.9%) and d6-dimethyl sulfoxide (DMSO-d6, 99.9%) were obtained from Novachem, Melbourne, Australia.

Methanol-Carbonate Solution Preparation

Immunohistochemical Analysis of GFAP in Intestinal Tissues

Sections of jejuna and colon were cut from paraffin blocks at 4 μm thickness and mounted onto silane-coated slides (Lomb Scientific Pty Ltd). Tissue samples were dewaxed in xylene (Merck Pty Ltd) and rehydrated through a graded series of ethanol (Chem Supply). Sections were immersed in 10 mM citrate buffer (pH 6.0, Sigma-Aldrich Pty Ltd) and antigen retrieval performed by heating sections in a microwave (LG) on high (800 W) until boiling and then on low (160 W) for 10 min. Sections were allowed to cool and endogenous peroxidise activity subsequently blocked with 3 % H 2 O 2 in methanol (Chem Supply). Non-specific antibody binding was blocked with 20 % normal horse serum (NHS) (Sigma-Aldrich Pty Ltd) in PBS for 30 min at room temperature (RT). Avidin-Biotin Blocking Kit (Vector Laboratories) was used to block endogenous avidin-biotin activity. Sections were incubated overnight with a primary goat polyclonal antibody (diluted in 10 % NHS) directed at GFAP at 1:400 (Santa Cruz Biotechnology, #SC-6171) at 4 °C. Sections were then incubated with an appropriate secondary antibody (20 min at RT), ABC labelling reagent (30 min at RT) and developed with 3,3′-Diaminobenzidine. Sections were counterstained with Lillie-Mayer's hematoxylin (HDS Scientific Supplies Pty Ltd), dehydrated and cleared in xylene (Merck Pty Ltd) before being mounted.

Sintered glass used as the membrane substrate was acquired from Ningbo Ja-Hely Technology Co., Ltd., Ningbo, China. This was in the form of flat circular discs of 25 mm diameter, 2 mm thickness and G5 porosity grade.

Analytical Reagent Preparation for Drug Analysis
