Genetically Modified TRPV1 Sensory Neurons

C57BL/6 mice were obtained from Jackson laboratories (Bar Harbor, ME). We generated mice expressing Ai32 (CHr2-eYFP) in TRPV1 sensory neurons (TRPV1-Ai32) by crossing homozygous Ai32 mice (B6;129S-Gt(ROSA)26Sortm32(CAG-COP4*H134R/EYFP)Hze/J) with homozygous TRPV1-cre mice (B6.129-Trpv1tm1(cre)Bbm/J) both of which were purchased from Jackson laboratories. All experiments were performed using 7-to 12-week-old male mice. Mice were housed in microisolator cages and fed irradiated food and acidified water. The University of Pittsburgh Institutional Animal Care and Use Committee approved all mouse protocols.

TRPV1-Cre or B6.129-TRPV1tm1(cre)Bbm/J (Stock number 017769); Rosa-LSL-tdTomato or B6.129S-Gt(ROSA)tm66.1(CAG-tdTomato)Hze/J or Ai9 (Stock number 005975); Rosa-LSL-GCaMP6f or B6J.Cg-Gt(ROSA)26Sortm95.1(CAG-GCaMP6f) Hze/MwarJ or Ai95D (Stock number 028865) strains were purchased from Jackson Laboratories (Cavanaugh et al., 2011 (link); Madisen et al., 2015 (link); Madisen et al., 2010 (link)). Calca-creERT2 was generously provided by Dr. Pao-Tien Chuang (Cardiovascular Research Institute, University of California, San Francisco, CA) (Song et al., 2012 (link)). Mice of both sexes were used in all experiments. Genotyping was performed according to information provided by Jackson Laboratories.

Optogenetic Manipulation of TRPV1 Neurons

All testing procedures were performed as per protocols approved by the Institutional Animal Care and Use Committee, and all efforts were made to minimize animal suffering in accordance with policies and recommendations established by the International Association for the Study of Pain. Homozygous B6.Cg-Tg(Thy1-COP4/EYFP)18Gfng/J (THY1-ChR2-YFP, Stock No: 007612), B6.129-Trpv1 tm1(cre)Bbm /J (TRPV1cre, stock no: 017769) and B6.Cg-Gt(ROSA)26Sor tm32(CAG-COP4 * H134R/EYFP)Hze /J (Ai32, stock no: 012569) mice were purchased from The Jackson Laboratory. An in-house breeding program was used to generate TRPV1-ChR2-EYFP mice by crossing the TRPV1cre driver mouse line with Ai32 conditional allele mice. Only male mice, aged 6-12 weeks old were used in all experiments. Animals were housed at a maximum of five per cage (30 3 20 3 15 cm) with ad libitum access to food and water. They were kept in controlled temperature of 23 ± 1 C on a 12-hrlight/dark cycle (lights on at 7:00 a.m.). Experiments were performed between 10 a.m. and 4 p.m. Different cohorts of mice were used for each test, except when stated.
