Cloning and Virus Production of Mouse Erg

Full-length mouse Erg was cloned into pEC3.1 which contains a CMV promoter and an IRES-Gfp flanked by attL sites. This vector was then recombined using the Gateway Cloning system into pAdBlockIt (Invitrogen). Virus was produced in HEK 293A cells (Invitrogen) following the manufacturer’s protocol. Virus was concentrated by pelleting the cells and resuspending in 1 ml of media before freeze thawing according to the manufacturer’s protocol. Virus was titered by infecting 293A cells with serial dilutions of viral supernatant. After 48 hours, the number of GFP-positive cells was counted and concentration calculated based on dilution. Control virus contained an additional Gfp in place of Erg.
