Antioxidant Biomarkers in ICR Mice

The animal experiment was approved by the Experimental Animal Care Ethics Committee of Northwest A&F University. A total of 320 ICR mice (male, 9 weeks old) with an average weight of 33 ± 2 g, SPF grade, were purchased from Vital River Laboratory Animal Technology Co., Ltd. (Beijing, China) (Approval Number: SCXK (Beijing) 2012-0001). Before the experiment, all experimental animals were bred adaptively in the animal room for one week. The conditions in the animal room were controlled as follows: the humidity was set to 55% ± 5% and the temperature was set to 25 ± 2 ℃, a 12-h circadian rhythm was ensured, and the experimental animals could drink water and eat freely. After all the mice had adapted to the environment, the experiment was started.
The aspartate aminotransferase (AST) kit, the alanine aminotransferase (ALT) kit, the total bilirubin (TBIL) kit, the alkaline phosphatase (ALP) kit, the total triglyceride (TC) kit, the total cholesterol (TG) kit, the total antioxidant capacity (T-AOC) kit, the catalase (CAT) kit, the glutathione (GSH) kit, the superoxide dismutase (SOD) kit, and the malonaldehyde (MDA) kit were purchased from Nanjing Jiancheng Bioengineering Institute (China).

Common Bile Duct Ligation Model in Mice

Male mice at 6-week age were intraperitoneally injected with chloral hydrate solution (0.3 g/kg). After anesthesia, a median abdominal incision was made and the common bile duct was found and carefully ligated with 7-0 Prolene (Ethicon, Somerville, NJ). The bile duct was dissected without ligation in sham operation, then wiped the incision with alcohol swab and injected with 0.5 mL of 0.9% saline on incision to improve recovery and survival after operation. Mice were kept warm at 37.5 C until recovery. Mice were then used for experiment 7 or 14 days after surgery; survival was monitored every day after surgery until 14 th day. Biomarkers for the liver functions of mice were tested with Alanine aminotransferase Assay Kit (C009-2-1), Aspartate aminotransferase Assay Kit (C010-2-1), Direct bilirubin (D-BIL) kit (C019-2-1), Total bilirubin (T-BIL) Kit (C019-1-1), and Alkaline phosphatase Assay kit (A059-2-2) from Nanjing Jiancheng Bioengineering Institute (Nanjing, China). Mice were tested with Toxin Sensor limuloid reagent detection kit (L00350, Genscript, Nanjing China) to exclude endotoxemia and liver infection 14 days after surgery.
