Analytical Standards for Chromatographic Analysis

Ultra-pure water (18 mΩ) was obtained from a Milli-Q water purification system (Millipore Co., Ltd., Milford, MA, USA). All stock solutions were sealed with Parafilm® and stored in a −20 °C freezer.

HepG2 Cell Lipotoxicity Modulation

Human hepatoma HepG2 cells were purchased from the Korean Cell Line Bank (KCLB, Korea) and cultured in Dulbecco's modified Eagle's medium (DMEM) supplemented with 10% fetal bovine serum (FBS), 1% penicillin, and streptomycin (Gibco, USA) in a humidified atmosphere under 5% CO2 at 37°C. HepG2 cells were seeded in 96-well plates. After reaching confluence, the cells were serum-starved overnight and exposed to 0.4 mM PA (Sigma-Aldrich, USA), in the presence or absence of 5 μM simvastatin (Sigma-Aldrich), 2 mM metformin (Sigma-Aldrich), 50 μM of hesperidin (LKT Laboratories, USA), narirutin (Sigma-Aldrich), nobiletin (Sigma-Aldrich), sinensetin (Sigma-Aldrich), or tangeretin (Sigma-Aldrich) for 24 h. The cells treated with simvastatin or metformin were used as the positive control.

BACE1 Inhibition Assay for Nobiletin, Tangeretin, and Resveratrol

Comprehensive Chemical Analysis of Ganpu Tea

HPLC Analysis of Citrus Flavonoids

Secondary Metabolite Profiling of Plants

Six independent plants were used as biological replicates, and about five leaves were sampled from each plant in secondary metabolic profiling. The secondary metabolic profiling was performed by LC-QTOF-MS using a modified method according to Yun et al. [37 (link)]. 100 mg freeze-dried powder was extracted with 80% methanol over night at 4°C. The mixture was centrifuged and filtered. Then, the metabolic profiling were performed using a QTOF 6520 mass spectrometer (Agilent Technologies, Palo Alto, CA, USA) coupled to a 1200 series Rapid Resolution HPLC system.
The raw data was processed by Agilent Mass Hunter Qualitative Analysis (version B. 04.00, Aglient Technologies) and Mass Profiler Software (version B.02.02, Aglient Technologies). Then PCA analysis was performed using the filtered and normalized data. Metabolite identification was carried out by comparing mass spectra and retention time with those of authentic standards. Nine authentic standards, namely, narirutin, naringin, hesperidin, neohesperidin, didymin, sinensetin, limonin, nomilin, and nobiletin were obtained from Sigma Chemical Co. (St. Louis, MO).
